摘    要:目的 探讨心纳素与人类心包间皮细胞之间的关系。 方法 分离人心包间皮细胞进行体外培养 ,用Fluo 3作为钙的指示剂 ,利用激光共聚焦显微镜测定 10 - 9mol L ,10 - 1 0 mol L ,10 - 1 1 mol L心钠素 (ANP)分别用作用于人心包间皮细胞后细胞内Ca2 (Ca2 )浓度变化。 结果 人心包间皮细胞存在细胞内Ca2 浓度的改变 ,随着ANP浓度的增加 ,细胞内Ca2 浓度出现非常明显的变化 (P <0 0 0 0 1) ;细胞内Ca2 浓度随时间变化也出现明显的变化 (P <0 0 0 0 1) ;不同时间测定的钙离子浓度与ANP浓度之间存在着交互作用 (P <0 0 0 0 1)。 结论 人心包间皮细胞存在细胞内Ca2 的改变 ;不同浓度的ANP作用于间皮细胞后引起细胞内Ca2 浓度出现明显不同的改变

EFFECT OF DIFFERENT KINDS OF ANP CONCENTRATION ON INTRACELLULAR CALCIUM LEVEL IN CULTURED HUMAN MESOTHELIAL CELLS
Abstract:Objective To investigate the Ca 2 changes in the pericardium mesothlial cells by confocal laser scanning microscopy(CLSM) and uncover the relationship between the ANP and the pericardium mesothelial cells. Methods Using Fluo 3 as a marker, Ca 2 in the cultured mesothelial cells were determined at one second interval by CLSM after 10 -9 ?mol/L,10 -10 ?mol/L,10 -11 ?mol/L atrial uretic peptide(ANP) being added, respectively. Results Calcium concentration in the mesothelial cells all was high after the three kinds of ANP were added. The calcium concentration of ANP 10 -9 ?mol/L cells elevated suddenly(about 10 times of the normal level, P <0 0001) and soon lowered beyond the normal level when the ANP appeared, the calcium concentration of ANP 10 -10 ?mol/L cells first had a light decreasing and then increased to 6 times higher than the normal level in the plasma ( P <0 0001), while the ANP 10 -11 ?mol/L group cells just displayed a 4 times increase and then continued ( P <0 0001). Conclusion Calciumion in cultured human mesothelial cells were changed after the stimulation of different kinds of ANP concentration. Different ANP concentrations could introduce different degree of calcium influx.
