Box-Behnken响应面法优化厚朴温中汤提取工艺

目的 优选与厚朴温中汤基准样品质量标准接近的提取工艺。方法 以橙皮苷、甘草酸、乔松素、厚朴酚的含量和出膏率作为关键质量属性，以单因素试验的结果作为基础，采用响应面试验法优化厚朴温中汤提取工艺。结果 厚朴温中汤最优提取工艺为加水780 mL，浸泡30 min，提取30 min，提取3次。此工艺条件下所得指标性成分的质量分数分别是0.97%、0.60%、0.15%、0.25%，出膏率为24.1%，与基准样品相接近。结论 厚朴温中汤提取工艺经过响应面法优化后更加可靠，符合基准样品质量标准范围，可为后续现代制剂的研究提供参考。

Optimization of Extraction of Houpo Wenzhong Decoction by Box-Behnken Response Surface Methodology

Objective To optimize the extraction close to the quality standards of reference samples of Houpo Wenzhong Decoction.Methods The extraction process of Houpo Wenzhong Decoction was optimized by response surface methodology with the content of hesperidin, glycyrrhizin, pinocembrin, and magnolol and extract yield as key quality attributes based on the results of single-factor experiments.Results The optimal extraction conditions of Houpo Wenzhong Decoction consisted of water addition volume of 780 mL, soaking time of 30 min, extraction time of 30 min, and extraction for three times. The content of index components obtained under the extraction conditions was 0.97%, 0.60%, 0.15% and 0.25% respectively, and the extract yield was 24.1%, close to the reference samples.Conclusion After optimization by the response surface methodology, the extraction process of Houpo Wenzhong Decoction is reliable and meets the quality standards of reference samples. This study is expected to provide support for the research on the subsequent development of modern preparations.
