异丙酚对缺氧复氧大鼠海马胶质细胞凋亡及Bcl-2和Bax蛋白表达的影响

摘    要:目的:观察异丙酚对缺氧复氧大鼠海马胶质细胞凋亡及Bc l-2和Bax蛋白表达的影响。方法:取新生2~3 dW istar大鼠海马星形胶质细胞,原代纯化培养3周。将细胞分为正常对照组,乳剂对照组,缺氧4 h后复氧12h、24 h、36 h、48 h、60 h、72 h组,加异丙酚250μmol/L缺氧4 h后复氧12 h、24 h、36 h、48 h、60 h、72 h组。分别于各时间点检测每组细胞凋亡及Bc l-2和Bax蛋白表达变化。结果:与正常对照组比较,缺氧复氧组细胞随着时间延长凋亡不断增加,Bc l-2蛋白24 h时间点表达最高,随后逐渐降低,Bax蛋白24 h表达低,随着时间延长不断增加,72 h时间点最高,Bc l-2/Bax蛋白比值于24 h、48 h和72 h时间点分别为1.4、0.8和0.6。与缺氧复氧组比较,异丙酚处理组凋亡明显减少,Bax蛋白表达降低,Bc l-2蛋白24 h内表达升高达峰值,此后有所降低但仍维持较高水平,且Bc l-2蛋白表达持续高于Bax蛋白,Bc l-2/Bax蛋白比值保持在1.6~1.8之间。结论:异丙酚可抑制缺氧复氧后海马反应性星形胶质细胞表达Bc l-2和Bax蛋白的动态变化,使Bc l-2/Bax蛋白比值持续保持较高水平而发挥良好的保护作用。
