大黄素对糖尿病大鼠肾脏结缔组织生长因子表达的影响

摘    要:目的研究大黄素对糖尿病大鼠肾脏结缔组织生长因子(CTGF)表达的影响,探讨大黄素肾脏保护作用的机制。方法 SD雄性大鼠54只,随机分为正常对照组(N组)、糖尿病对照组(D组)、糖尿病大黄素治疗组(E组)。由大鼠尾静脉单次注射链脲佐菌素(60mg/kg)制备糖尿病大鼠模型。经大黄素治疗糖尿病大鼠2,4,8周后,分别观察其对大鼠血糖、24 h尿蛋白定量和肾皮质CTGF表达的影响。结果与正常对照组相比,糖尿病大鼠24 h尿蛋白定量及肾组织CTGF表达显著增加(P均0.01);经大黄素治疗8周后,糖尿病大鼠的24 h尿蛋白定量和CTGF表达减少(P均0.05)。结论糖尿病大鼠肾组织CTGF表达增加,大黄素可能通过下调糖尿病大鼠肾脏CTGF表达而达到肾脏保护作用。

Abstract:Objective It is to observe the influence of emodin on connective tissue growth factor(CTGF) in the renal tissue of diabetic rats,and to explore the mechanism of emodin in the renal protective effect.Methods 54 male SD rats were randomly divided into three groups: normal control group(group N),diabetic control group(group D),diabetic rats treated with emodin group(100 mg/(kg·d))(group E).The diabetic rat models were induced by a single injection of streptozotocin(60 mg/kg) via coccygeal vein.The levels of blood glucose,excretion of 24 h urinary protein and Connective Tissue growth factor(CTGF) in the renal tissue were measured after 2,4,8 weeks of treatment.Results The levels of 24 h urinary protein and the expressions of CTGF in group D and E were significantly increased than those in group N(P<0.01).After treated with emodin for 8 weeks,these indexes were decreased in group E(P<0.05).Conclusion The expression of CTGF is increased in the renal tissue of diabetic rats.Emodin has some renal protective effects in diabetic rats,partly through down-regulating the CTGF expression.
