HSP70在胃癌中的表达及意义

目的:探讨热休克蛋白70(HSP70)及其mRNA在胃癌、癌旁组织、慢性胃炎和正常胃黏膜组织中的表达情况及与临床、病理资料之间的关系。方法:采用组织芯片技术、免疫组织化学方法检测病理诊断明确的60例胃癌组织标本及对应的距胃癌组织边缘1.5cm处的癌旁组织、距胃癌组织边缘5cm以上的正常组织以及胃炎组织中HSP70的表达差异,RT-PCR法对10例患者的胃癌组织及对应的癌旁组织、慢性胃炎组织和正常组织的HSP70mRNA的表达情况进行检测,结合临床病理资料进行统计分析。结果:HSP70的表达:在60例胃癌组织中的阳性表达率为75%,明显高于癌旁组织(P<0.01)、胃炎组织(P<0.01)和正常组织(P<0.01);在mRNA水平,胃癌组织的HSP70表达也明显高于其余3组(P<0.001)。HSP70的表达与患者年龄、性别、肿瘤组织类型和浸润胃壁深度无明显相关性(P>0.05);与肿瘤分化程度、远处转移和临床病理分期具有相关性(P<0.05)。结论:胃癌中HSP70的表达水平明显高于癌旁组织、胃炎组织和正常组织,提示与胃癌发生、发展密切相关;而且与患者年龄、性别、肿瘤组织类型和浸润胃壁深度无明显相关性;与肿瘤分化程度、远处转移和临床病理分期具有相关性。

HSP70 Expression in Gastric Carcinoma and Its Significance

Objective:To investigate the expression of heat shock protein 70(HSP70) and mRNA in gastric cancer,cancer,chronic gastritis and normal mucosa tissues,and the relationship between the clinical and pathological data.Methods:Using tissue microarray technology,immunohistochemistry to detect the definitive histological diagnosis of 60 cases of gastric carcinoma specimens from the gastric tissue and the corresponding edge 1.5cm cancer tissue from gastric carcinoma,edge 5cm above normal tissue and chronic gastritis in the expression difference of HSP70,RT-PCR in 10 cases of patients with gastric carcinoma and the corresponding cancer,chronic gastritis and normal tissues and the expression of HSP70mRNA was detected,in combination with the clinical pathological data for statistical analysis.Results:The expression of HSP70:in 60 cases of gastric carcinoma of the positive expression rate of 75%,significantly higher than adjacent tissues(P<0.01),chronic gastritis(P<0.01) and normal tissues(P<0.01);in the level of mRNA,HSP70 expression in gastric cancer tissue was significantly higher than the other 3 groups(P<0.001).The expression of HSP 70 and patient’ age,gender,tumor invasion depth of gastric tissue type and no significant correlation(P>0.05);and the degree of tumor differentiation,distant metastasis and the clinical pathological staging a correlation(P<0.05).Conclusion:The expression of HSP70 in gastric cancer was significantly higher than cancer,gastritis and normal tissues of gastric cancer,tips,and development are closely related;and patient’ age,gender,tumor invasion depth of gastric tissue type and no significant correlation;and tumor differentiation degree,distant metastasis and the clinical pathological staging with correlation.
