伊贝沙坦对糖尿病大鼠心肌组织葡萄糖转运蛋白4表达的干预研究

摘    要:目的观察伊贝沙坦对糖尿病大鼠心肌病模型心肌肥大和心肌细胞葡萄糖转运蛋白4(GLUT4)表达的影响。方法将Wistar大鼠随机分为A组(健康对照组)、B组(糖尿病对照组)、C组(伊贝沙坦治疗组),D组(小剂量胰岛素治疗组)和E组(小剂量胰岛素+伊贝沙坦治疗组)。将B组、C组、D组和E组大鼠制成糖尿病模型后,C组和E组予以50 mg/(kg.d)-1伊贝沙坦灌胃,同时D组和E组予以普通胰岛素2.0u Bid皮下注射。观察第11周时,大鼠的心体比值(心脏重量/体重的比值)及相关的血糖变化。免疫组化方法观察各组大鼠心肌细胞GLUT4蛋白表达的情况。结果①B组、C组、D组和E组大鼠心体比值较A组均明显增高(P〈0.05),E组心体比值较B组、C组和D组减少(P〈0.05),而C组心体比值较B组和D组减少(P〈0.05);②B组、C组、D组和E组血糖较A组明显增高(P〈0.01),E组血糖较B组、C组和D组降低(P〈0.05),C组血糖较B、D组降低(P〈0.05),而后两组之间的差别无统计学意义;③免疫组化法半定量分析显示,E组心肌细胞表达的葡萄糖转运蛋白4(GLUT4)较B组、C组和D组增高(P〈0.05),C组GLUT4蛋白较B组和D组增高,B组和D组之间无明显差别,而此4组GLUT4蛋白的表达较A组均明显减少(P〈0.01)。结论糖尿病大鼠早期应用伊贝沙坦可增加心肌细胞GLUT4蛋白的表达,抑制糖尿病心肌细胞的肥大和组织重构,并有轻度的降低血糖的作用。而以上这些作用,是独立于胰岛素之外的。

Study on Intervention of Epistan for expression_of GLUT4 in Diabetic Rat Myocardiac Cell

Abstract:Objective To observe the effect of hypertrophy and GLUT4 expression of myocardiac cell of diabetic rat.Methods The rats were divided into 5 groups,group A(health control),group B(diabetics),group C(epistan),group D(insulin) and group E(epistan and insulin).The diabetic model was established and then group C and E were lavaged with epistan.Group D and E were injected with insulin.The H/B ratio,glucose change and some other were observed after 11week.The GLUT4 expression was observed.Results ①H/B ratio was higher in group B,C,D and E than that in group A,H/B in group E was less than those in group B,D and C,H/B in group C was less than those in group B and D.②Blood glucose was lower in group B,C,D and E than that in group A,in group E was less than those in group B,D and C,blood glucose in group C was less than those in group B and D.③GLUT4 expression of GLUT4 in was lower in group B,C,D and E than that in group A,in group E was higher than those in group B,D and C,in group C was higher than those in group B and D.Conclusion Epistan might improve the in early stage of diabetic rat so that to inhibit hypertrophy and reconstruction of myocardiac cell and decrease blood glucose except independent insulin function.
