消斑通脉合剂对血管平滑肌细胞增殖和P21waf1/cip1蛋白表达的影响

摘    要:目的:探讨中药复方消斑通脉合荆药物血清对血小板源性生长因子(PDGF—BB)诱导的大鼠血管平滑肌细胞(VSMC)增殖与细胞周期分布的影响及相关作用机制。方法:体外培养VSMCs,PDGF—BB刺激VsMCs,采用MTr法检测药物血清对VSMCs增殖的影响;流式细胞仪分析各组细胞周期分布情况;应用Western印迹法检测药物血清对VSMcs中P21waf1/cip1蛋白的表达变化。结果:MTT法显示药物血清呈剂量依赖性抑制VSMCs增值,高剂量与低剂量比较有显著差异(P〈0.05);流式细胞仪分析结果显示与模型组比较,低剂量与高剂量组明显提高G0/G1期百分率(P〈0.01)、减少s期百分率(P〈0.01),低剂量与高剂量组比较有显著差异(P〈0.05);Western印迹法显示与模型组比较,高剂量与低剂量可明显提高VSMCs中P21waf1/cip1蛋白表达(P〈0.01),低剂量与高剂量比较有统计学差异(P〈0.01)。结论:中药复方消斑通脉合剂药物血清通过促进VSMCs中P21waf1/cip1。蛋白表达,阻滞VsMcs细胞周期进程,起到抑制VsMcs增殖作用。

Abstract:Objective : To study the anti - proliferation and cell cycle blocking effects of the xiao ban tong mai he ji on rat vascular smooth muscle cells(VSMCs) in vitro. Methods: VSMCs were treated with various concentration of the medi- cal serum of xiao ban tong mai he ji for 24 hours. The cell viability was detected by MIT method. Flow cytometry was used to detect the cell cycle. Western blotting was performed to detect the expression of cell cycle regulatory p21 protein. Results : High concentration of the medical serum inhibited the proliferation of VSMCs. Compared with the control group, the expression of P21 protein in the high concentration group were significantly decreased. Conclusion :The medical serum of xiao ban tong mai he ji inhibit of the proliferation and blocks cell cycle progression of VSMCs. These effects are as- sociated with the expression of cell P21 protein.
