血竭加味颗粒对自然衰老子宫肌瘤模型小鼠IGF-Ⅰ表达的影响

摘    要:目的:建立自然衰老和青年小鼠子宫肌瘤模型,研究血竭加味颗粒对子宫肌瘤模型小鼠子宫组织中胰岛素样因子Ⅰ(IGF-Ⅰ)的表达。方法:采用雌激素负荷法建立子宫肌瘤模型。将40只11月龄自然衰老小鼠、50只2~3月龄青年小鼠ICR雌性小鼠各随机分成5组,分别为模型组、对照组、血竭加味颗粒组、米非司酮组、桂枝茯苓胶囊组,通过观察子宫系数、子宫组织中IGF-Ⅰ的表达比较各组差异。结果:(1)血竭加味颗粒、米非司酮、桂枝茯苓胶囊均对雌激素负荷子宫肌瘤模型小鼠均有明显抑瘤作用。(2)血竭加味颗粒、米非司酮对自然衰老小鼠子宫组织IGF-Ⅰ的表达有显著抑制作用(P〈0.01),桂枝茯苓胶囊对IGF-Ⅰ表达的抑制作用相对较小(P〈0.05)。结论:血竭加味颗粒抑制子宫组织中IGF-Ⅰ的阳性表达可能为抑制子宫肌瘤的机制之一。

Abstract:Objective:To study the effect of Xuejiejiawei granule on Insulin-lile Growth Factor-I(IGF-Ⅰ)on natural aging and young mouse models of uterine hysteromyoma.Methods:Uterine hysteromyoma models were established with estrogen load.Then forty eleven-month-old natural aging and fifty two to three-month-old young ICR female mouse were randomly divided into five groups,as a model group,control group,Xuejiejiawei granule group,mifepristone group ,Guizhi Fuling capsule group,uterine coefficients and expressions of IGF-I in uterine tissue were observed and compared among groups.Results:(1) Xuejiejiawei Granule,mifepristone and Guizhi Fuling capsule inhibited obviously the grouth of uterine hysteromyoma of mouse modles with estrogen load. (2) Xuejiejiawei Granule and mifepristone significantly inhibited expressions of IGF-I in uterine tissue of natural aging mouse(P〈0.01),while the inhibitiones of GuizhiFuling capsule on IGF-Ⅰwere relatively small (P〈0.05). Conclusion:Xuejiejiawei Granule inhibits expression of IGF-Ⅰin uterine tissue of mouse may one of the mechanisms inhibits the growth of uterine hysteromyoma.
