红花黄色素对单侧输尿管梗阻大鼠模型肾间质纤维化的影响

摘    要:目的:探讨红花黄色素对大鼠肾间质纤维化的影响。方法:将40只SD雄性大鼠随机分为假手术(SO)组、单侧输尿管梗阻(UUO) 组、血管紧张素Ⅱ受体拮抗剂厄贝沙坦组(IRB组) 和红花黄色素组(SY组) 。大鼠肾组织行苏木精-伊红和Masson染色,计算肾间质纤维化的相对面积。免疫组化技术检测I型胶原(Col I)和纤维连接蛋白(FN)在肾组织中的表达。结果:SY治疗组和IRB治疗组能有效改善UUO大鼠的肾脏间质纤维化,SY组和IRB组中各时间点Col I和FN蛋白的表达均低于UUO组(P0.05)。结论:红花黄色素可减轻UUO大鼠肾间质纤维化程度,延缓肾脏间质纤维化。

Abstract:Objective:To investigate the antifibrotic effect of safflor yellow on renal intersititium in unilateral ureteral obstruction (UUO) rats.Methods:40 male Sprague-Dawley rats were randomly divided into sham-operated group (SO group), unilateral ureteral obstruction model group (UUO group), irbesartan group (IRB group) and safflor yellow group (SY group). The area of renal interstitial fibrosis was calculated by HE and Masson staining, while the expression of collagen I (Col I) and fibronectin (FN) were detected by immunohistochemistry. Results:Renal interstitial fibrosis was improved significantly in both IRB group and SY group. The expression levels of Col I and FN were significantly reduced in the IRB and SY group compared with UUO group, notwithstanding the lowest in the SO group. Conclusion:Safflor yellow could ameliorate renal interstitial fibrosis which might be realized by reducing the exressio n of Col I and FN, it could delay the progress of renal interstital fibrosis in UUO rat model.
