Abstract:Objective To investigate the effect of long-chain non-coding RNA(LncRNA)Down syndrome cell adhesion molecule antisensei(DSCAM-AS1)targeting miR-627-3p on the proliferation and invasion ability of human skin squamous cell carcinoma and the underlying molecular mechanism.Methods The expression of and miR-627-3p in human immortalized epidermal cells HaCaT and three types of skin squamous cell carcinoma cells(SCC13,A431 and HSC-5)were detected by real-time fluorescence quantitative PCR(RT-qPCR).The DSCAM-AS1 small interfering RNA and miR-627-3p mimics were transfected into A431 cells,respectively.The cell proliferation rate was detected by the CCK-8 method.Cell migration and invasion were detected by transwell assay,and CyclinD1,MMP2 and MMP9 protein expression was detected by Western blotting.Luciferase analysis was performed to examine the targeting regulation of miR-627-3p by DSCAM-AS1.Western blotting was used to detect the ability of DSCAM-AS1 and miR-627-3p to regulate PI3 K/AKT signaling pathway.Results The expression of DSC4M-AS1 in the three types of skin squamous cell carcinoma cells was significantly higher than in HaCaT cells,while the expression of miR-627-3p)in the three types of skin squamous cell carcinoma cells was significantly lower than in HaCaT cells(P<0.05).After interfering with DSCAM-AS1 or over-expressing miR-627-3p,the expression of CyclinDl,MMP2 and MMP9 protein in A431 cells was significantly reduced,and the cell proliferation rate,cell migration and invasion numbers were significantly reduced(P<0.05).DSCAM-AS1 targets and negatively regulates miR-627-3p expression.Compared with the interference of DSCAM-AS1 expression,the expression of CyclinD1,MMP2 and MMP9 proteins in A431 cells were significantly increased,and the cell proliferation rate,cell migration and invasion numbers were significantly increased after interfering with DSCAM-AS1 and rniR-621-3p(P<0.05).After interfering with DSCAM-AS1,the expression of p-PI3 K and p-AKT proteins in A431 cells was significantly reduced(P<0.05).Compared with interference with DSCAM-AS1,the expression of p-PI3 K and p-AKT protein in A431 cells was significantly increased after interfering with DSCAM-ASl and miR-627-3p(P<0.05).Conclusion Interference with DSCAM-AS1 inhibits the PI3 K/AKT signaling pathway by targeting miR-621-3p,thereby inhibiting the proliferation,invasion,and migration of skin squamous carcinoma cells.
