兔腰椎融合术中BMP和bFGF对骨髓间充质干细胞的作用

摘    要:目的评价骨形态发生蛋白(BMP)和碱性成纤维细胞生长因子(bFGF)诱导的兔骨髓间充质干细胞(MSC)复合多孔生物陶瓷对提高腰椎融合率的作用。方法将32只家兔随机分为4组(n=8),用下列移植物分别进行腰椎后外侧路融合:Ⅰ组单独应用MSC;Ⅱ组以bFGF作用MSC;Ⅲ组用BMP-2作用MSC;Ⅳ组用BMP-2和bFGF共同作用MSC。实验动物术后6周处死,利用放射学、手工生物力学和组织学方法观察和分析椎体融合的情况。结果影像学和手工力学检测,Ⅳ组椎体融合情况要比其它3组好得多;组织学检测示,在Ⅳ组中,可以看到在融合区周围出现大量的新生成的骨小梁。Ⅱ,Ⅲ组在融合区新生骨小梁较Ⅳ组少,而是新生骨小梁与纤维结缔组织混杂出现;Ⅰ组新生骨小梁显著减少,生物陶瓷微孔内多为纤维结缔组织。结论经BMP和bFGF体外培养的MSC可取得比单独应用MSC更好的融合效果。
