肝细胞癌中上皮钙黏蛋白、尿激酶型纤溶酶原激活物及其受体的表达及意义

摘    要:
目的 研究上皮钙黏蛋白 (E- CD)、尿激酶型纤溶酶原激活物 (u PA )及其受体 (u PAR)与肝细胞癌(HCC)的关系及其在 HCC侵袭转移中的作用。 方法 分别应用免疫组织化学、原位杂交方法检测 E- CD和u PA、u PAR m RNA在 5 2例 HCC、4 2例肝硬化组织和 2 3例正常肝组织中的表达。 结果 在 HCC中 ,E- CD的表达明显减低 ,u PA、u PAR表达明显增高。在 HCC中 ,E- CD在转移组的表达明显低于无转移组 ,E- CD的表达与病理学分级有关 ,分化程度越高 ,E- CD水平越低 ;u PA、u PAR在转移组的表达明显高于无转移组 ,且其表达水平的增高与包膜侵犯有关。相关分析显示 E- CD与 u PA、u PAR呈负相关。 结论  E- CD和 u PA、u PAR在 HCC中的表达明显异常 ,E- CD表达减低和 u PA、u PAR表达增高与 HCC侵袭转移密切相关

Abstract:
Objective To study the relationship between E- cadherin( E- CD) ,urokinase- type plas- mingen activator( u PA) ,u PA receptor( u PAR) and tumor cell invasion,metastasis in hepatocellular carci- noma( HCC) . Methods The expression of E- CD by immunohistochemistry and the expressions of u PA, u PAR by in situ hybridization were detected in5 2 cases of HCC,42 cases of liver cirrhosis and2 3cases of normal livertissue. Results Compare with theirexpression in livercirrhosis and normal liver tissue,the expressions of E- CD and u PA,u PAR in HCC were abnormal:E- CD expression was reduced,u PA,u PAR were overexpression significantly. In HCC,E- CD expression with invasion was reduced significantly. The expressions of u PA,u PAR with invasion were increased significantly,the overexpressions of u PA, u PAR were associated with capsule invasion. E- CD showed negative correlation with u PA,u PAR. Conclusion Expressions of E- CD and u PA,u PAR were abnormal in HCC. That the reduced expression of E- CD and overexpressions of u PA,u PAR contribute significantly to HCC invasion and metastasis.
