阿托伐他汀对心肌梗死后大鼠主动脉平滑肌NADPH氧化酶的影响

摘    要:目的探讨心肌梗死后大鼠主动脉平滑肌NADPH氧化酶的变化及阿托伐他汀的干预作用。方法取雄性SD大鼠,通过是否结扎左前降支分为手术组和假手术组,手术组大鼠于术后第二天随机给予阿托伐他汀[10mg/(kg.d)](心梗干预组)或等体积生理盐水(心梗组)灌胃共5周,第6周处死大鼠取胸主动脉平滑肌,采用WST-1法及RT-PCR法分别测定平滑肌超氧阴离子(O2-)浓度及Nox1 mRNA和Nox4 mRNA表达,并探讨其相关性。结果心梗组O2-浓度、Nox1 mRNA及Nox4 mRNA表达高于假手术组(P<0.01)和心梗干预组(P<0.05)。O2-浓度分别与Nox1 mRNA及Nox4 mRNA表达呈正相关(r=0.53,P<0.05;r=0.56,P<0.05)。结论心梗后大鼠平滑肌NADPH氧化酶系统被激活,阿托伐他汀通过降低Nox1 mRNA及Nox4 mRNA表达而减少O2-产生。

Influence of atorvsatin on NADPH oxidase of aortic smooth muscle in rat after myocardial infarction

Abstract:Objective To explore the change of NADPH oxidese and the intervention effect of atorvsatin on NADPH oxidase of atorvsatin aortic smooth muscle in rats after myocardial infarction(MI).Methods Male Sprague-Dawley rats were divided into operated group and sham group according to ligation or not of anterior descending coronary artery.The operated rats were assigned randomly to intragastrically receive atorvastatin[10 mg/(kg·day)](MI intervention group)or saline(MI group) respectively on the second day after operation for 5 weeks.On the sixth week,the rats were killed and aortic smooth muscle was taken.The concentration of superoxide(O-2) was measured with WST-1 method and the expressions of Nox1 mRNA and Nox4 mRNA were detected with RT-PCR,and the correlations between the concentration of O-2 and the expression of Nox1 mRNA and Nox4 mRNA were respectively calculated.Results The concentration of O-2 and the expression of Nox1 mRNA and Nox4 mRNA in MI group increased significantly compared to sham group(P<0.01) and MI intervention group(P<0.05).The concentration of O-2 positively correlated to the expression of Nox1 mRNA and Nox4 mRNA(r=0.53,P<0.05;r=0.56,P<0.05 respectively).Conclusions The NADPH oxidase system is activated in aortic smooth muscle of rats after MI.Atorvsatin decreases the production of O-2 by inhibiting the expression of Nox1 mRNA and Nox4 mRNA.
