兔面神经撞击伤后生长相关蛋白、神经丝蛋白变化及丙酸睾丸酮促修复作用

摘    要:目的 探讨兔面神经撞击伤后生长相关蛋白 (GAP - 4 3 )、神经丝蛋白 (NFP2 0 0 )的变化及丙酸睾丸酮 (TP)的促修复作用。方法 将 84只兔分为等渗盐水对照组和TP治疗组 ,用生物撞击机致面神经干中度损伤。采用免疫组化和Western印迹等方法观察GAP - 4 3、NFP2 0 0 的变化。结果 伤后两组面神经核内GAP - 4 3免疫反应阳性神经元数量和含量迅速增加 ,并于伤后 2周达高峰 ,TP治疗组显著高于对照组 ;伤后 7天 ,NFP2 0 0 免疫反应阳性神经元和含量降至最低 ,对照组更为明显。伤后早期神经纤维变性、髓鞘崩解 ,伤后 2周治疗组可见少量新生神经纤维。结论 GAP- 4 3和NFP2 0 0 的表达与神经的再生修复过程密切相关 ,TP具有显著的促面神经损伤修复作用。

Changes of GAP-43 and NFP_(200 ) after Facial Nerve Impact Injury and The Promotive Effects of TP on Nerve Regeneration in Rabbits

Abstract:Objective To study the changes of growth associated protein (GAP-43) and neurofilament protein (NFP 200 ) in facial nerve nucleus, as well as the promotive effects of testosterone propionate (TP) after injury. Methods Eighty-four rabbits were randomly divided into the control group (saline treatment group) and the TP treatment group. Moderate injuries of right facial nerve were made with BIM-II impact machine. The changes of GAP-43 and NFP 200 were observed using immunohistochemistry and Western blot methods. Results The number of neuron with positive GAP-43 immuno-reaction and the content of GAP-43 in facial nerve nucleus increased rapidly after injury, and reached its peak value 2 weeks after trauma. These elevations were more significant in the TP group. The number of the neuron with positive NFP 200 immuno-reaction and the content of NFP 200 in facial nerve nucleus fell to the lowest value 7 days after facial nerve injury, and they were more remarkable in the control group. Pathological examination showed the degeneration of nerve fibers and disintegration at medulla sheath of the early stage after trauma. Two weeks after injury, Newborn nerve fibers were found in the TP group. Conclusions The expressions of GAP-43 and NFP 200 were well correlated with the regeneration of injured nerve. TP might exert markedly promotive effects on the recovering process of nerve injury.
