Toll样受体TLR-2,TLR-4和TLR-9在皮肤颜面粟粒性狼疮皮损中的表达

摘    要:目的检测Toll样受体(TLR)-2,4和9在颜面粟粒性狼疮(LMF)皮损中的表达,探讨其在LMF发病中的作用。方法选用20例LMF,15例银屑病患者皮损处皮肤以及10例正常皮肤的石蜡切片,用免疫组化的方法检测TLR-2,4和9的表达情况。结果 TLR-2在正常皮肤、银屑病及LMF均呈高表达,比较差异均无统计学意义(P>0.05);TLR-4在正常皮肤、银屑病呈低或无表达,而LMF呈中等表达,比较差异均有统计学意义(P<0.05);TLR-9在正常皮肤为低表达,而LMF及银屑病呈高表达,与正常皮肤比较差异有统计学意义(P<0.05)。结论 TLR-2,TLR-4和TLR-9在LMF皮损中均存在表达,共同参与了LMF发病过程中的免疫应答及调节,其中TLR-4和TLR-9的表达高于正常皮肤,提示其在LMF发病过程中可能起着主要作用。

Abstract:Objective To investigate the expression of TLRs-2,4 and 9 in the lesions of lupus miliaris faciei and explore the possible role in the pathogenesis of lupus miliaris faciei. Methods Tissue specimens from 20 patients with lupus miliaris faciei, 15 patients with psoriasis, and 10 with normal skins were used to detect the expression of TLR-2, 4 and 9. Results TLR-2 was detected mainly in the upper layer of epidermis of both lupus miliaris faciei and psoriasis lesions, while it was detected in the whole epidermis except the basal layer in normal skins, the differences of expression had not significant (P>0.05). In lupus miliaris,the expression of TLR-4 was much higher than that in the normal skins or psoriatic lesions (P<0.05),the expression of TLR-9 was higher than that of normal skins(P<0.05) ,while it was similar to psoriatic lesions. Conclusion TLR-2, 4 and 9 were involved in the immune response and modulation in the pathogenesis of lupus miliaris faciei. The expressions of TLR-4 and TLR-9 were much higher than that of the normal skins, suggesting that they may play more important roles in the immune response of lupus miliaris faciei.
