蓝萼甲素对宫颈癌HeLa细胞的作用及其相关机制研究

摘    要:目的观察蓝萼甲素对宫颈癌HeLa细胞的增殖抑制作用,探讨其诱导凋亡的机制。方法采用体外细胞培养方法,以不同浓度蓝萼甲素作用于HeLa细胞,用四甲基偶氮唑盐法(MTT法)检测细胞的存活率;透射电镜观察细胞的形态学变化;流式细胞仪(AnnexinV-FITC)检测细胞凋亡率的变化;Western blot检测凋亡相关蛋白Bcl-2、Bax、Caspase-3、Caspase-9蛋白的表达情况。结果与对照组相比,蓝萼甲素对HeLa细胞有增殖抑制作用,呈剂量、时间依赖趋势;Bcl-2蛋白表达下降,Bax蛋白表达上调,Caspase-3、Caspase-9蛋白表达上调。结论蓝萼甲素对宫颈癌HeLa细胞有增殖抑制及诱导凋亡的作用,其分子机制可能与线粒体信号通路有关。

Abstract:Aim To investigate the effects of Glaucocalyxin A on proliferation on HeLa cell,and the mechanism of Glaucocalyxin A induced apoptosis.Methods Using vitro cell culture methods,HeLa cell were treated by different concentrations of Glaucocalyxin A,after different times of treatment,the cell viability was measured by MTT method.Morphological changes were observed by electron micrograph.Apoptosis of HeLa cells after 48 h of different concentrations of Glaucocalyxin A exposure was detected by flow cytometry(AnnexinV-FITC staining);the protein expressions of Bax,Bcl-2,Caspase-3,Caspase-9 were assayed by Western blot.Results Compared with the control group,Glaucocalyxin A had a significant anti-proliferation effect on HeLa cells in a dose and time-dependent manner.The protein of bax was upregulated,and bcl-2 was downregulated.Caspase 3 was upregulated,caspase 9 was upregulated.Conclusions Glaucocalyxin A can inhibit the proliferation of HeLa cells,inducing HeLa cell to apoptosis remarkably and concentration-dependently.The mitochondrial signal transduction pathway is involved in its mechanism.
