MMP/TIMP表达失衡在肾细胞癌中的研究

目的应用组织芯片技术探讨基质金属蛋白酶-2（MMP-2）、基质金属蛋白酶-7（MMP-7）和金属蛋白酶抑制因子-2（TIMP-2）在肾细胞癌中的表达情况及相关性。方法采用免疫组化法（SP法）检测82例肾细胞癌和40例正常对照者中MMP-2、MMP-7和TIMP-2的表达情况。结果 MMP-2在肾细胞癌组中表达阳性率71.9%,高于对照组15.0%,MMP-7在肾细胞癌组中表达阳性率67.1%,高于对照组20.0%,并随组织学分级、淋巴结转移、临床分期增高表达增强（P〈0.05）;TIMP-2在肾细胞癌组中表达阳性率34.1%,略高于对照组30.0%,无统计学意义（P〉0.05）,并随组织学分级、淋巴结转移和临床分期增加表达降低（P〈0.05）;在肾细胞癌组中两两之间呈负相关。结论 MMP-2-7/TIMP-2失衡可能参与了肾细胞癌的发生发展过程,联合表达可用于判断肾细胞癌的恶性程度。

The study of the expression imbalance of MMP/TIMP in renal cell carcinoma by tissue microarrays

Objective To investigate the expression and correlation among MMP-2、MMP-7 and TIMP-2 in renal cell carcinoma(RCC) by tissue microarrays.Methods The RCC tissue microarrays containing 82 cases specimen were used to determine MMP-2、MMP-7and TIMP-2 expression by immunohistochemistry(SP).Results The expression rate of MMP-2 was 71.9% in RCC,which was higher than in control group(15.0%),and MMP-7 was 67.1% in RCC higher than in control group(20.0%).The expression of MMP-2/-7 were higher in RCC than in control group,and was prominently expressed within the histology grades,metastasis of lymph nodes and clinical phases(P < 0.05).The expression rate of TIMP-2 was 34.1% in RCC,which was higher than in control group(30.0%),but no significant correlation with control group(P > 0.05).The expression of TIMP-2 was reduced within the histology grades,metastasis of lymph nodes and clinical phases(P < 0.05).There was negatively relation between MMP-2/-7 and TIMP-2 in RCC.Conclusion MMP-2-7/TIMP-2 may play an important role in the development of RCC.The combined expression may evaluate the malignant degree of RCC.
