| 摘 要： | 目的：培养鉴定骨髓间充质干细胞（MSCs），检测survivin的表达。方法：应用Ficoll法分离培养MSCs，流式细胞仪检测其特异性表面标志物，RT—PCR检测survivin mRNA表达。结果：培养MSCs的超微结构显示了干细胞特性，流式细胞仪检测证实为MSCs，survivin mRNA表达阳性。结论：Survivin在正常人骨髓间充质干细胞中有表达，提示它可能参与了其生物学行为并影响间充质干细胞存活。 |

| Abstract: | Objective：To culture and identify the normal adult bone mesenehymal stem cells （MSCs）, and to detect the expression of survivin in them in vitro and discuss its significance. Methods： MSCs isolated by Ficoll - Hypaque density gradient method were cultured in the LG - DMEM with 15% FBS. The specific surface markers were detected by flow cytometry, and the expression of survivin mRNA was determined by RT - PCR. Results： The ultramierostructure of MSCs identified by flow cytometry showed the characteristics of stem cells. The expression of survivin mRNA in MSCs was positive. Conclusion： Survivin expression is positive in the normal adult MSCs, indicating that survivin may play an important role in biological behaviour of MSCs and affect MSC survival. |
