血管瘤中血管内皮生长因子表达的定量研究

摘    要:目的观察血管内皮生长因子(VEGF)在血管瘤中的表达,并探讨VEGF在血管瘤中的表达水平与DNA倍体、细胞增殖活性及血管瘤侵袭的关系.方法应用流式细胞仪及免疫荧光技术检测几种类型的血管瘤组织中VEGF表达量荧光指数FI、DNA含量(DNA指数,DI)及细胞增殖活性(增殖指数,PI).结果在血管瘤中多数为DNA异倍体、各类血管瘤FI、DI、PI值之间差异无显著性意义(P>0.05);包膜完整组与无包膜或包膜不完整组之间FI值差异有显著性意义(P<0.01);异倍体与二倍体之间FI值差异有显著性意义(P<0.01);VEGF表达阳性组与VEGF表达阴性组之间PI值差异有显著性意义(P<0.01).结论VEGF表达水平与血管瘤的侵袭性、DNA倍体及细胞增殖活性密切相关.提示阻止VEGF表达能作为治疗血管瘤的方法.

Abstract:Objective To observe vascular endothelial growth factor(VEGF) expression in hemangioma and the relationship between VEGF expression and DNA ploidy,cellular proliferation activity and invasion of hemangioma. Methods Expression of VEGF (fluores cence index, FI), DNA content (DNA index, DI), cellular proliferation activity (proliferation index, PI) in hemangioma were detected using flow cytometry and immunofluorescence staining technique. Results Majority of hemangioma were heteroploid. There was no significant difference in FI, DI, PI ofeach type of hemangioma ( P >0.05) .There were great significant differences between the intact capsule group and incomplete capsule group ( P <0.01) in FI, between DNA aneupliod group and DNA diploid group ( P <0.01) in PI, and between VEGF expression positive group and VEGF expression negative group ( P <0.01) in PI.Conclusion Expression of VEGF is highly related to the invasion, DNA ploidy and cellular proliferation activity of hemangioma, which suggests that inhibition of VEGF expression can be used in the treatment of hemangioma.
