高效液相色谱法测定川葛颗粒中葛根素的含量

摘    要:目的建立高效液相色谱法测定川葛颗粒中葛根素含量。方法色谱柱Zorbax Eclipes XDB(150 mm×4.6 mm,5μm);流动相甲醇∶水(25∶75);流速1.0 ml/min;检测波长250 nm;柱温25℃。结果葛根素在0.090 4~0.904 0μg范围内线性关系良好(r=0.999 9,n=5);平均回收率为97.48%(n=9),RSD为1.47%。结论本方法操作简单、准确可靠,可用于川葛颗粒的质量控制。

Abstract:Objective To establish a high performance liquid chromatography(HPLC) method for determination of puerarin in Chuange granules. Methods Separation of samples was achieved on a Zorbax Eclipes XDB column(150 mm × 4.6 mm,5 μm).The mobile phase consisted of methanol-water(25:75) at a flow rate of 1.0 ml/min.The detection wavelength was 250 nm and the column temperature was 25℃. Results The standard curves for puerarin were linear in the ranges of 0.090 4-0.904 0 μg(r =0.999 9,n =5);The average recovery rate was 97.48%(n =9),RSD was 1.47%. Conclusion The method is fast,simple,specific,reliable,and can be used for quality control of Chuange granules.
