地龙、乌梢蛇对系膜增生性肾炎大鼠诱导型一氧化氮合酶、内皮素-1表达的影响

摘    要:目的观察地龙、乌梢蛇对系膜增生性肾炎(MsPGN)大鼠诱导型一氧化氮合酶(iN—OS)、内皮素-1(ET-1)表达的影响,探讨其治疗作用及机制。方法用改良慢性血清病法制备MsPGN大鼠模型,以地龙、乌梢蛇高低剂量灌胃,设正常对照组及模型组,治疗8周后测血白蛋白(ALB)、肌酐(Scr)、尿素氮(BUN),观察肾脏病理变化及iNOS、ET-1表达。结果与正常对照组比较,模型组24h尿蛋白、Scr及BUN升高(P〈0.01)而ALB降低(P〈0.01);与模型组比较,各治疗组24h尿蛋白、Scr及BUN降低(P〈0.05)且ALB升高(P〈0.01),降24h尿蛋白和升ALB作用地龙高剂量组优于低剂量组(P〈0.05)。iNOS、ET-1表达水平模型组高于正常对照组(P〈0.01),各治疗组低于模型组(P〈0.01)。结论地龙、乌梢蛇均能降低MsPGN大鼠蛋白尿,升高ALB,地龙高剂量优于低剂量,乌梢蛇高、低剂量差异无统计学意义(P〉0.05),二药均可降低Scr、BUN,其机制可能与抑制iNOS、ET一1在肾组织表达有关。

Abstract:Objective To observe the impacts of geosaurus and zaocys on the expressions of induc- ible nitric oxide synthase ( iNOS ) and endothelin - 1 ( ET - 1 ) expression in the rats with mesaugio proliferative glomerulonephritis(MsPGN) and explore the therapeutic effects and the mechanism. Methods The modified chronic serum sickness method was adopted to prepare MsPGN rat model. The geosaurus and zaocys of the high and low doses were used for the lavage in the rats. A normal group and a model group were set up. 8 weeks later, blood albumin ( ALB ), serum ereatinine (Scr) and urea nitrogen ( BUN ) levels were detected. The renal pathological changes and the expressions of iNOS and ET - 1 were observed. Results Compared with the normal group, the levels of 24 h urine protein, Scr and BUN were increased and ALB level was reduced in the model group(P 〈0.01 ). Compared with the model group,the levels of 24 h urine protein,Scr and BUN were all reduced in all the treatment groups ( P 〈 0.05 ) and ALB level was increased ( P 〈 0.01 ). The de- crease of 24h urine protein level and the increase of ALB level in the geosaurus - high - dose group were su- perior to that in the low - dose group( P 〈 0.05 ). The expressions iNOS and ET - 1 in the model group were significantly higher than those in the normal group ( P 〈 0.01 ), and the results in all the treatment groups were lower than those in the model group ( P 〈 0.01 ). Conclusion Geosaurus and zaocys all reduce urine protein level and increase ALB level in MsPGN rats. The results in the geosaurus - high - dose group are bet- ter than those in the low - dosage group. There is no statistical significant difference between the zaocys - high - dose group and the low - dose group ( P 〉 O. 05 ). Either geosaurus or zaocys can reduce the levels of Scr and BUN and the mechanism may be related to the inhibition of the expressions of iNOS and ET - 1 in the renal tissues.
