胶艾汤止血机制的实验研究

摘    要:
目的探讨胶艾汤的止血作用机制。方法将SD大鼠分为正常对照组,胶艾汤低、中、高剂量组,宫血宁组。分别给予0.9%氯化钠溶液,胶艾汤8.57、17.14、34.28 g.kg-1(按成人临床1、2、4倍剂量给药),宫血宁0.6 g.kg-1(按成人临床6倍剂量给药),灌胃给药,1次.d-1,连续用药7 d,心脏穿刺取血,测定凝血酶原时间(PT)、部分凝血活酶时间(APTT)及血栓素B2(TXB2)、6-酮-前列腺素F1a(6-keto-PGF1a)的水平。结果胶艾汤中、高剂量明显缩短大鼠的PT、APTT,显著提高TXB2而下调6-keto-PGF1a的水平,与正常对照组比较,差异有统计学意义(P<0.05或P<0.01)。结论胶艾汤可能是通过激活内源性、外源性凝血系统和调节TXB2、6-keto-PGF1a的水平发挥止血功能。

Experimental Research of Jiaoai Decoction on Hemostasis Mechanism

Abstract:
Objective To explore the hemostasis mechanism of jiaoai decoction. Methods SD rats were divided into a control group,a low- dose jiaoai decoction group ,a medium- dose jiaoai decoction group, a high - dose jiaoai decoction group and gongxuening group. They were treated with 0.9% NaC1 solu- tion,jiaoai decoction 8.57,17.14 and 34.28 g- kg-1 (based on the clinical dosages 1,2 and 4 times as same as the adults)and gongxuening 0.6 g ~ kg-1 (based on the clinical dosage 6 times as same as the a- dults) separately. The gastric infusion was adopted for the medication, once per day, continuously for 7days. The blood was collected by cardiac puncture for the detection of prothrombin time (PT) and active partial thromboplastin time(AFFF) as well as the levels of thromboxane B: ( TXB2 ) and 6 - ketone - prostaglandin Fj, (6 - keto - PGF1a ). Results Jiaoai decoction of the medium dose and high dose shortened apparently PT and APTT and improved significantly TXB2 level and down - regulated 6 - keto - PGF1a. The statistical sig- nificant differences presented as compared with the normal control group( P 〈 0.05 or P 〈 0.01 ). Conclusion Jiaoai decoction plays hemostasis function probably through activating the endogenous and exogenous coag- ulation system and regulating the levels of TXB2 and 6 - keto - PGF1a
