胃癌组织MMP-2、MMP-9、Shh蛋白的表达及与临床病理特征的关系

摘    要:目的 探讨胃癌组织基质金属蛋白酶-2(MMP-2)、MMP-9及音猬因子(Shh)蛋白的表达及与临床病理特征的关系。方法 收集笔者医院54例胃癌组织与癌旁组织石蜡标本,应用免疫组化法检测MMP-2、MMP-9、Shh蛋白的表达,并分析与临床病理特征的关系。结果 胃癌组织中的MMP-2、MMP-9、Shh蛋白阳性表达比例明显高于癌旁组织(P<0.05)。MMP-2与MMP-9表达呈正相关(r=0.633),MMP-2与Shh表达呈正相关(r=0.413),MMP-9与Shh表达呈正相关(r=0.527)。胃癌组织MMP-2蛋白的表达与TNM分期、淋巴转移有关(P<0.05);MMP-9蛋白的表达与TNM分期、组织类型、淋巴转移有关(P<0.05);Shh蛋白的表达与TNM分期有关(P<0.05)。结论 MMP-2、MMP-9、Shh蛋白在胃癌组织中表达均上调,三者可能相互协同或调解对肿瘤的分化、侵袭与转移起作用。

Abstract:Objective To explore the xpression of matrix metalloproteinase-2 (MMP-2), MMP-9 and sonic hedgehog (Shh) protein in gastric cancer tissues and the relationship with clinicopathological features. Methods Paraffin specimens of 54 cases of gastric cancer and adjacent tissues were collected,and immunohistochemical staining was used to detect the expression of MMP-2, MMP-9 and Shh proteins. Results The positive rates of MMP-2, MMP-9 and Shh protein in gastric cancer tissues were significantly higher than those in adjacent tissues (P<0.05).The expression of MMP-2 was positively correlated with the expression of MMP-9 (r=0.633), and the expression of MMP-2 was positively correlated with the expression of Shh (r=0.413), and the expression of MMP-9 was positively correlated with the expression of Shh (r=0.527).The expression of MMP-2 protein in gastric carcinoma was related to TNM stage and lymph node metastasis (P<0.05).The expression of MMP-9 protein was related to TNM staging, tissue type and lymph node metastasis (P<0.05).The expression of Shh protein was related to TNM staging (P<0.05). Conclusion The expression of MMP-2, MMP-9 and Shh protein were up-regulated in gastric carcinoma,and the three may play a role in tumor differentiation, invasion and metastasis cooperate or mediate each other.
