乳癌组织中胸苷磷酸化酶检测

摘    要:目的:研究乳癌组织中胸苷磷酸化酶(TP)蛋白和mRNA的表达与微血管密度(MVD)的关系及其临床意义。方法:采用RT-PCR技术检测30例乳癌和17例正常乳腺组织中TP mRNA的表达水平。用免疫组化法检测82例乳癌(包括上述30例)和17例正常乳腺组织TP和CD34的表达,并计算MVD。结果:乳癌细胞的TP蛋白和TP mRNA的表达水平明显高于正常乳腺组织(P〈0.05);TP蛋白和TP mRNA的高表达率与乳癌组织学分级有关(P〈0.05);乳癌组织的MVD值明显高于正常乳腺组织(P〈0.01);TP蛋白高表达组MVD明显高于TP低表达组(P〈0.01);乳癌TP mRNA高低表达组之间的MVD差异无统计学意义(P〉0.05)。结论:TP蛋白和TP mRNA在乳癌组织中表达增强,其表达与乳癌的组织学分级有关。

Detection of thymidine phosphorylase expression in breast cancer tissue

Abstract:Aim: To study the relationship between the expressions of TP, TP mRNA and microvessel density(MVD) in breast cancer. Methods: The mRNA expression of TP in 30 cases of breast carcinoma and 17 cases of normal breast tissue was measured using RT-PCR.The expressions of TP and CD34 in sections of 82 cases of breast carcinoma and 17 cases of normal breast tissue were detected using immunohistochemistry. MVD was calculated according the SP staining results. Results: The levels of TP and TP mRNA expression in breast carcinoma tissue were significantly higher than those in normal breast tissue (P<0.05); the levels of TP and TP mRNA expression were correlated with histological grades in breast cancer tissue(P<0.05);MVD in breast cancer tissue was higher than that in normal breast tissue (P<0.01); MVD in TP-higher-expression group was significantly higher than that in TP-lower-expression group (P<0.01); MVD had no significant difference between groups with different TP mRNA expression levels (P>0.05). Conclusion:Higher expression of TP and TP mRNA in breast cancer tissue is obseved and related to histological grades.
