摘    要:目的观察以嘉陵江(重庆段)为水源的管网末梢水中有机提取物对雄性生殖系统的影响.方法用固相萃取技术(吸附剂:GDX-102树脂)富集水中有机物(OE).昆明种雄性小鼠随机分为5组,分别用DMSO(溶剂对照,0.005 L/kg)、OE 12.5 L/kg、OE 25 L/kg、OE 50 L/kg、CP(阳性对照,40 mg/kg)连续腹腔注射5 d,第15天处死动物取材.应用放射免疫法(RIA)检测血清中的睾酮(T),硫代巴比妥酸法(TBA)检测睾丸组织液中丙二醛(MDA).光镜下计数畸形精子.光镜和电镜观察睾丸病理组织学改变.结果与DMSO相比,OE组睾酮水平显著性降低;MDA的含量未见显著差异(P>0.05);同时畸形精子数显著增加.光镜下发现生精细胞疏松脱落,数量减少.电镜下可见间质细胞、支持细胞、生精细胞超微结构的改变.结论以嘉陵江(重庆段)为水源的管网末梢水中的有机提取物对小鼠具有一定的雄性生殖毒性.

Abstract:Objective To study the toxicity of the organic extract of tap water on the male reproductive system in mice. Methods Organic pollutants were extracted from tap water with GDX-102 resin and the extract was injected into the peritoneal cavity of male mice in the dosages of 12.5, 25 and 50 L/kg daily for consecutive 5 days. DMSO served as the solvent control and cyclophosphamide as the positive control. At day 10 after the last injection, the mice were killed and serum level of testosterone was determined with radioimmunoassay (RIA). The MDA content of the testis homogenate was measured with thiobarbituric acid method (TBA). The number and deformity of the sperms were observed with optic microscopy. The sections of the testis were prepared with the routine procedure and the histopathological changes were studied with optic and eletronic microscopies. Results When the data of the mice of the organic extract group were compared with those of the DMSO control group, it was found that the organic extract of tap water resulted in a significant decrease of serum testosterone and a significant increase of deformed sperms. No significant difference between the MDA content of the mice of 2 groups was shown. Optic microscopy revealed the detachment of the spermatogenic cells and decrease of the number of spermatid cells. The changes of ultramicrostructure of the leydig cells, sertoli cells and spermatogenic cells were found with electron microscopy. Conclusion The findings of our study suggest that the organic pollutants of tap water result in damages of the reproductive system of male mice.
