Klotho基因多态性与冠心病及海南长寿人群的相关性研究

目的 探讨Klotho基因多态性与冠心病及海南长寿人群的相关性.方法 分别随机选取入户调查的海南省百岁老人50例(长寿组)、在301医院海南分院住院的冠心病患者50例(冠心病组)和在健康体检中心体检的健康人群50例(对照组),记录入选的150例对象的性别、年龄、体质指数(BMI)及血压、血糖、血脂等生化指标,并运用ASP-PCR技术检测Klotho基因G-395A、C1818T、F352V、C370S四个位点.结果 冠心病组、长寿组、对照组Klotho基因的C1818T位点均为T纯合子;冠心病组及对照组Klotho基因的F352V位点均为F纯合子、C370S位点均为C纯合子;冠心病组、长寿组、对照组Klotho基因的G-395A位点基因型频率及等位基因频率组间比较差异均无统计学意义(P>0.05).结论 Klotho基因G-395A、C1818T、F352V、C370S多态性可能与冠心病、海南长寿人群均无相关性.

Correlation Research between Klotho Gene Polymorphisms with Coronary Heart Diseases and Longevity in Hain-an

Objective To investigate the correlation of Klotho gene polymorphisms with coronary heart diseases and longevity in Hainan .Methods Totally 50 cases of longevity were randomly selected from household survey of cente-narians , and 50 patients with coronary heart diseases in 301 hospital of hainan branch institute were selected as experimen-tal group, and 50 healthy people who received physical examination in physical examination center were selected as control group.The sex, age, body mass index (BMI) and blood pressure, blood sugar, blood lipids and other biochemical indi-cators were recorded.ASP-PCR was used to detect G -395A, C1818T, F352V and C370S sites of Klotho gene.Results The C1818T site of Klotho gene was T homozygous in all 3 groups.The F352V site was F homozygous, and the C370S site was C homozygous in both coronary heart disease group and health control group .There was no significant difference a-mong coronary heart disease group , longevity group and control group regarding genotype and allele frequency in G -395A site of Klotho gene (P>0.05).Conclusion There is no correlation among polymorphism of G -395A, C1818T, F352V and C370S of Klotho gene, coronary heart disease and longevity in Hainan Province .
