摘    要:目的:采用HPLC法测定盐酸芬氟拉明血药浓度,对5只家兔进行药代动力学研究,并就一例芬氟拉明中毒致死者的血、尿和脏器检材的毒物检测进行了分析。方法:流动相为甲醇-002mol·L-1庚烷磺酸钠(25∶75),二苯胺为内标,在268nm进行检测。结果:芬氟拉明线性范围为5~200μg·L-1(r=09986),最低检测浓度5ng,平均方法回收率为977%。日内、日间RSD均小于6%,达峰时间为(15±3)h,峰浓度(654±49)μg·L-1,药时曲线下面积为(1754±328)μg·L-1·h-1。结论:此方法稳定、可靠,可用于血药浓度分析和法医中毒检测。

Abstract:bjective: A HPLC method was established for determining fenfluramine in rabbit serum. The study devoted to pharmacokinetic parameters for five rabbits and has been applied to determine the concentration of fenfluramine in blood, urine and organs of postmortem autopsy. Methods: Methanol-sodium heplanesulphonate(2575)was selected as mobile phase and diphenylamine was used as achieved by internal standard. The detective wavelength was 268 nm. Results: The ranges of the calibration curves were 5200 gL-1(r=09986) and detection limits were 5 ng,while the average recovery of the method was 977%.The RSD for intra-day and inter-day assays were all less than 6%. tmax were (153)h and Cmax were (65449)gL-1. AUC were (1754328)ghL-1. Conclusion: This method is stable and reliable. The techniquAuthor e was useful for the analysis of fenfluramine in serum and the determination of this toxicant in forensic practice.
