纤维素酶法提取翠云草总黄酮的工艺研究

摘    要:目的研究纤维素酶法提取翠云草总黄酮的最佳提取工艺。方法以翠云草总黄酮含量为指标,分别研究了酶用量、酶解温度、pH值和酶解时间对总黄酮提取率的影响,在此基础上设计了L9(34)正交实验,确定最佳提取工艺。结果纤维素酶法提取翠云草中总黄酮的最佳工艺条件为:酶用量为22 U/g,酶解温度为45℃,pH值为5.0,酶解时间为60min。结论优化后的提取工艺条件合理可行,为翠云草总黄酮的工业化生产提供理论参考依据。

Abstract:ObjectiveTo investigate the optimal extraction technology of total flavonoids from Selaginella uncinata(Desv.) Spring with cellulase. MethodsUsing the content of flavonoids as the estimative targets,the effects of four factors of cellulose concentration,hydrolysis temperature,pH and hydrolysis time on the extraction were investigated.On the basis as above,L9(34) orthogonal experiment was designed to optimize the extraction technology of Selaginella uncinata(Desv.) Spring. ResultsThe optimum conditions of cellulase enzymatic extraction of total flavonoids with cellulase from Selaginella uncinata(Desv.) Spring were as follows: concentration was 22U/g,hydrolysis temperature was 45℃,pH was 5.0,hydrolysis time was 60min. ConclusionThe optimum extraction conditions of cellulase enzymatic extraction are feasible,which can be taken as further theoretical basis for industrial production of the total flavonoids from Selaginella uncinata(Desv.) Spring.
