巴戟天多糖含药血清对体外成骨细胞DKK-1表达的影响

摘    要:目的探讨巴戟天多糖含药血清对成骨细胞增殖、分化及DKK-1蛋白表达的影响。方法源于大鼠颅盖骨的原代成骨细胞中加入不同浓度的巴戟天多糖含药血清进行干预。MTT法观察巴戟天多糖含药血清对成骨细胞增殖的影响,用硝基苯磷酸盐(P-nitrophenyl phosphate,PNPP)偶氮法观察含药血清对成骨细胞碱性磷酸酶(alkaline phosphatase,ALP)活性的影响,用蛋白印记法观察含药血清对成骨细胞DKK-1蛋白表达的影响。结果巴戟天多糖含药血清可使体外培养成骨细胞的增殖率明显提高(P<0.01),并能提高成骨细胞ALP活性(P<0.01)。此外,巴戟天多糖含药血清可以下调成骨细胞DKK-1蛋白的表达。结论巴戟天多糖含药血清可促进成骨细胞的增殖能力和ALP活性,并可通过下调成骨细胞DKK-1蛋白的表达影响骨代谢。

Abstract:ObjectiveTo investigate the effects of drug-containing serum of Morinda officinalis polysaccharide on the proliferation,differentiation and DKK-1 in osteoblast(OB). MethodsDrug-containing serum of Morinda officinalis polysaccharide with different contcentrations was fed to calvaria bone specimens obtained from rats.The proliferation of osteoblasts was analyzed by MTT assay,the ALP activity of OB was observed by PNPP and the expression of DKK-1 was measured by Western Blot. ResultsDrug-containing serum of Morinda officinalis polysaccharide had notable effects on the proliferation(P<0.01),promoted the activity of ALP(P<0.01) and inhibited the expression of DKK-1 in vitro cultured OB(P<0.01). ConclusionDrug-containing serum of Morinda officinalis polysaccharide can promote the proliferation and the activity of ALP,and significantly inhibit the expression of DKK-1 in vitro cultured OB.
