鬼臼毒素衍生物ZM对人骨肉瘤细胞HOS增殖及PCNA基因表达的影响

摘    要:
【目的】研究鬼臼毒素衍生物ZM对HOS细胞增殖及PCNA表达影响。【方法】培养HOS细胞,实验随机分为正常对照组、鬼臼毒素衍生物ZM组和阳性对照组。用MTT法检测鬼臼毒素衍生物ZM对HOS细胞的生长抑制作用,DNA Ladder凝胶电泳法分析鬼臼毒素衍生物ZM的诱导凋亡作用,RT-PCR检测PCNA基因的表达。【结果】与正常对照组相比,鬼臼毒素衍生物ZM能明显抑制HOS细胞的生长,且与浓度、时间呈依赖性,其IC50(48 h)为(1.32±0.3)μmol/L;鬼臼毒素衍生物ZM处理HOS细胞后,DNA凝胶电泳结果显示呈明显的梯状条带,RT-PCR结果显示PCNA表达显著下调。【结论】鬼臼毒素衍生物ZM对人骨肉瘤细胞HOS有明显的生长抑制作用,该作用可能与诱导凋亡和PCNA基因的表达下调有关。

Abstract:
【Objective】To study the effect of Podophyllotoxin ZM on proliferation and PCNA expression of human osteosarcoma cell line HOS.【Methods】Human osteosarcoma cell HOS were randomly divided into control,Podophyllotoxin ZM,and positive control group.The growth inhibition of Podophyllotoxin ZM on HOS was observed by MTT.Analysis of Podophyllotoxin ZM-induced apoptosis by DNA Ladder gel electrophoresis,Expression of PCNA was determined by RT-PCR.【Results】Compared with control group,Podophyllotoxin ZM significantly inhibited the growth of HOS cells in a dose and time dependent,The IC50(48 hours,1.32±0.3) μmol/ L;After treatment of Podophyllotoxin ZM on HOS cells,DNA gel electrophoresis results showed a clear laddering.RT-PCR showed down-regulation of PCNA expression.【Conclusion】The growth of human osteosarcoma cell line HOS significantly was inhibited by Podophyllotoxin ZM,and the effect was related to apoptosis and the down-regulation of PCNA expression.
