山羊卵泡大小对卵母细胞体外发育能力的影响

摘    要:
目的比较不同大小山羊卵泡的卵母细胞体外成熟、体外受精及胚胎发育的能力。方法收集不同大小山羊卵泡的卵母细胞,分3组进行体外培养。Ⅰ组:卵泡直径≥5.0mm;Ⅱ组:卵泡直径3.0~4.9mm;Ⅲ组:卵泡直径〈3.0mm。观察各组卵母细胞体外成熟率、体外受精率、卵裂率及囊胚形成率。结果3组卵母细胞体外成熟率与受精率差异无统计学意义(均P〉0.05)。Ⅲ组卵母细胞的卵裂率及囊胚形成率分别为53.8%和28.6%,均显著低于Ⅰ组和Ⅱ组(均P〈0.01)。结论在进行山羊未成熟卵体外成熟培养时,卵泡直径≥3mm的卵母细胞体外发育潜能较卵泡直径〈3mm的卵母细胞好。

Abstract:
Objective To compare in vitro maturation(IVM),in vitro fertilization(IVF) and embryos developmental capability of goat oocytes derived from follicles with different size. Methods The oocytes derived from follicles with different size were divided into three groups for in vitro culture. Group Ⅰ:follicular diameter size ≥5.0 mm. Group Ⅱ :follicular diameter size were 3.0 ~4.9 mm. Group Ⅲ:follicular diameter size 〈3.0 mm. Then the oocytes were cultured for observing rates IVM,IVF,cleavage and blastocyst. Results There was no significant difference among three groups in IVM and IVF rates (all P〉0.05). The cleavage and blastocyst rates of oocytes from Group Ⅲ were 53.8% and 28.6%,which were significantly lower than those from Group Ⅰ and Group Ⅱ(all P〈0.01). Conclusion For in vitro culture of goat immature oocytes,the development capability of oocytes derived from follicular diameter size ≥3 mm were better than those from follicular diameter size 〈3 mm.
