摘    要:目的研究干扰素γ(IFN-γ)对人肝癌细胞B7分子表达的影响。方法选用人肝癌HepG2细胞进行体外培养,经过不同浓度的IFN-γ干预后,采用MTT法检测细胞增殖作用,RT-PCR检测B7-1和B7-2mRNA的表达。结果不同浓度的IFN-γ对HepG2细胞体外生长有明显抑制作用;干预后HepG2细胞的B7基因表达明显增加。结论IFN-γ能直接抑制HepG2细胞增殖,显著上调HepG2细胞B7基因的表达,具有一定的抗肿瘤效应。

Abstract:Objective To study the effects of IFN-γ on expression of B7 molecules in human liver neoplasm cell line. Methods Human liver neoplasm HepG2 cells were cultured in vitro in the medium with IFN-γ respectively, cell proliferation effect was measured by MTT assay; expression of B7-1 and B7-2 mRNA was determined by RT-PCR. Results MTT shows that IFN-γ could reduce the HepG2 cell proliferation, expression of B7-1 and B7-2 significantly increased in the IFN-γ group. Conclusion IFN-3, can inhibit cell proliferation. IFN-γ markedly enhance the expression of B7-1 and B7-2 in HepG2 line and olavs a role in anti-cancer effect.
