维拉帕米对人晶状体上皮细胞整合素表达的抑制作用

摘    要:目的研究钙通道阻滞剂维拉帕米对人晶状体上皮细胞(LECs)株SRA01/04整合素表达的影响。方法对人LECs株SRA01/04进行培养并传代,分别以0.02、0.04、0.08mmol/L的维拉帕米作用于SRA01/0424h,经流式细胞仪检测SRA01/04各整合素的阳性表达率。结果整合素α2、α3、α5、β1、β2在人LECs株SRA01/04中呈阳性表达,维拉帕米对其表达具有抑制作用,并随药物浓度的增加而逐渐增强(P〈0.05)。结论钙通道阻滞剂维拉帕米对LECs整合素的表达具有抑制作用,可能阻碍整合素介导的LECs黏附、移行和增生,有望成为防治后发性白内障的有效药物。

Abstract:Objective To prevent the formation of after cataract is very important for the improvement of visual function after cataract surgery. Research showed that calcium channel blocker can inhibit the proliferation of lens epithelial cells by destroying the integrin. Present study was to explore the influence of verapamil,a calcium channel blocker,on integrins expression in cultured human lens epithelial cells(HLECs). Methods The human lens epithelial cells line,SRA01/04,was cultured in Dulbecco' s Modified Eagle Medium/Nutrient Mixture F-12 (DMEM/F-12) supplemented with 10% fetal bovine serum (FBS) and maintained at 37 ℃ in 5% CO2 atmosphere. Following trypsinisation, cells were incubated with 0.02,0. 04,0. 08 mmol/L verapamil for 24 hours, respectively. The cultured cells without verapamil were as control. The positive percentages of different integrin subunits in this HLECs line were detected by flow eytometry. Results The expression rate of integrin subunits α2 ,α3, α5, β1,β2 in cultured SRA01/04 line was significantly different among control groups and 0. 02 , 0. 04 , 0. 08 mmol/L verapamil groups (Xα2 2 =4034. ,Xα3^2 =3009. 1,Xα5^2 =6558.3,Xβ1^2 =1 849.8,Xβ2^2 =100.2,P 〈0.01).The expression ofα2,α3,α5,β1, β2 in cultured SRA01/04 line was gradually declined with the decrease of verapamil concentration and showing considerable differences in comparison with control group( P 〈 0.01 ) ,indicating a strong inhibition for the proliferation of HLECs in vitro in a dose-dependent manner(P 〈0. 01 ). Conclusion Verapamil inhibits integrin subunits expression in SRAO1/04 and therefore arrest the proliferation, differentiation,and migration of residual LECs after cataract surgery. Verapamil may be a suitable drug of preventing and treating posterior capsule opaeification (PCO).
