摘 要: | 目的:探讨Annexin A1在小鼠胚泡植入期的作用。方法:双向聚丙烯酰胺凝胶电泳(2D-PAGE)分析孕d3、d5、d7小鼠子宫内膜蛋白质组,用差异蛋白质组学显示pI约5.1、分子量约38kD的蛋白点在d3、d5上调,且在d5更明显。经基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)测定其胶内酶解后的肽质量指纹谱(peptide mass fingerprint,PMF)和数据库搜索对此蛋白点进行鉴定;再用RT-PCR、原位杂交、Western blot和免疫组织化学技术进行验证分析。结果:该蛋白点鉴定为鼠源性Annexin A1;d5小鼠子宫内膜组织中Annexin A1的mRNA和蛋白质水平均增加。结论:Annexin A1可能参与胚泡着床这一重要生命活动过程。

Expression of Annexin A1 in Endometrium during the Blastocyst Implantation in Mouse |

Abstract: | Objective: To investigate the role of Annexin A1 during the blastocyst implantation of mouse. Methods: Total proteins of mice endometria from pregnant d 3,d 5 and d 7 were analyzed by two-dimensional gel electrophoresis and image analysis,respectively. Differential expression protein spots were excised by dot-gel digestion with trypsin. Then peptide mass fingerprinting (PMF) was submitted to an appropriate software (Mascot: PMF) for identifying proteins. A differential protein spot was checked by RT-PCR,Western blotting,immunohistochemistry and in-situ hybridization. Results: The differential protein spot was identified as Annexin A1 by PMF. The expression level of Annexin A1 mRNA and protein reached the maximum at the d 5 of pregnant. Conclusion: Annexin A1 might play an important role during the blastocysti mplantation. |
