外源性氧自由基DPPH对家兔胸主动脉血管内皮舒张功能的影响

摘    要:目的:观察外源性自由基DPPH所致的家兔离体胸主动脉内皮依赖性舒张功能的影响。方法:采用不同浓度DPPH孵育家兔离体血管环,观察它对血管内皮依赖性舒张功能的影响;并在电镜下观察胸主动脉血管环组织形态结构的变化。结果:不同浓度的DPPH孵育后能明显抑制胸主动脉环对乙酰胆碱(Ach)诱导的血管内皮依赖性舒张反应,其中0.25μmol/L孵育家兔离体血管环30min损伤适中。电镜下可以看到正常对照组内皮结构完整,细胞排列紧密,细胞间连接紧密;0.25μmol/LDPPH损伤组可见内皮结构不完整,大量内皮细胞的脱落,崩解,可见细胞核崩解,细胞间连接松散,有炎性细胞浸润。结论:成功制备胸主动脉血管环氧化损伤模型,为研究保护血管内皮功能的药物提供前期基础。

Abstract:Objective: To observe the vascular endothelial dysfunction induced by 1,1 -dipheyl -2 -picrylhydrazyl (DPPH). Methods : An incubation with different concentrations of DPPH medium for different time could induce the impairment of the endothelium - dependent relaxation response (EDRR) to acetylcholine (Ach) in the isolated rabbit thoracic aortic rings. The pathomorphological changes in rabbit thoracic aortic rings were also observed by transmission electron microscope. Results: DPPH can significantly produced a decrease of EDRR induced by Ach. Conclusion: We successful prepare the model of the vascular endothelial dysfunction and provide research basic for vascular - protected drugs.
