摘    要:目的 探讨胃癌细胞中SerpinB5能与MAFbx的相互作用及其作用位点.方法 采用酵母双杂交及免疫共沉淀方法检测SerpinB5与MAFbx之间的相互作用.分别用RNA干扰及瞬间转染pGBKT7-SerpinB5调节SerpinB5表达水平.在胃癌细胞株SUN-16中研究SerpinB5对MAFbx表达的影响.用同源建模的方法构建出MAFbx的结构信息;并以分子对接的手段来预测SerpinB5与MAFbx相互作用的位点.结果 酵母双杂交及免疫共沉淀均证实SerpinB5能与MAFbx相互作用;MAFbx的表达水平随着SerpinB5的表达水平变化而变化.同源建模和分子对接的方法预测SerpinB5与MAFbx蛋白相互作用的关键残基是PR0261、ASN361和LYS362.结论 胃癌细胞中SerpinB5能与MAFbx相互作用,且SerpinB5上的PR0261、ASN361、LYS362残基是其与MAFbx蛋白相互作用的可能位点.

Abstract:Objective To explore the interaction between SerpinB5 and MAFbx in gastric cancer cell and to identify the interaction sites.Methods The interaction between SerpinB5 and MAFbx was screened and validated by yeast two-hybrid screening and co-immunoprecipitation.The expression of MAFbx was analyzed after SerpinB5 expression being modified by RNA interference and pGBKT7-SerpinB5 transfection.The impact of SerpinB5 on the expression of MAFbx was studied in gastric cancer cell line SUN-16.A model of MAFbx was constructed by homology modeling.The related residues for interaction were analyzed by Autodock4.0.Results The interaction between SerpinB5 and MAFbx was validated.The expression of MAFbx changed along with SerpinB5 expression.Amino acids including PRO261,ASN361,and LYS362 were key residue in the interaction of SerpinB5 and MAFbx.Conclusion SerpinB5 interacts with MAFbx in gastric cancer cell.Amino acids including PRO261,ASN361,and LYS362 are potential binding sites.
