一测多评法测定黄连中小檗碱、巴马汀、黄连碱、表小檗碱、药根碱含量

摘    要: 目的在建立黄连中的5个主要生物碱同步测定方法的基础上,建立一测多评法的方法学考察模式,验证该方法在黄连中应用的可行性和技术适应性。方法以黄连药材为研究对象,以药材中5个典型生物碱为指标,建立小檗碱与巴马汀、黄连碱、表小檗碱、药根碱的相对校正因子,并用该校正因子进行巴马汀、黄连碱、表小檗碱、药根碱的含量计算(计算法),实现一测多评;同时采用外标法测定药材中该5种生物碱的含量(实测法),并比较计算值与实测值的差异,以验证一测多评法的准确性和可行性。结果28批黄连药材和饮片中5个生物碱的含量可以用一测多评法进行测定,其计算值与实测值间无显著差异(RSD<5%)。结论以小檗碱、巴马汀、黄连碱、表小檗碱、药根碱同步测定的一测多评法控制黄连药材和饮片的质量是可行的、准确的。

Abstract:OBJECTIVE To establish a new method of quality evaluation and validate its feasibilities by the simultaneous quantitative assay of five alkaloids in rhizome of Coptis chinensis.METHODS The new quality evaluation method,quantitative analysis of multi-components by single marker(QAMS),was established and validated with rhizoma of Coptis chinensis.Five main alkaloids,berberine,palmatine,coptisine,epiberberine and jatrorrhizine,were selected as analytes to evaluate the quality.Their contents in 28 batches of samples,collected from different areas,were determined by both external standard method and QAMS.The method was evaluated by comparison of the quantitative results between external standard method and QAMS.RESULTS No significant differences were found in the quantitative results of five alkaloids in 28 batches of Coptis chinensis determined by external standard method and QAMS(RSD<5%).CONCLUSION It is feasible and suitable to evaluate the quality of rhizome of Coptis chinensis by QAMS.
