血管抑素对糖尿病大鼠视网膜组织变化及其VEGF的差异表达

摘    要:目的研究血管抑素体内抑制视网膜新生血管形成疗效及对血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法建立链脲佐菌素糖尿病大鼠模型,分A组(造模实验组),B组(造模对照组),每组36只,取正常SD大鼠36只作为C组(正常对照组)。予以玻璃体腔内注射血管抑素后观察视网膜HE染色结果及大鼠的视网膜VEGF的表达。结果 A、B组大鼠的血糖值明显高于同期C组;A组即注射血管抑素与B组即注射磷酸盐缓冲液(phosphate buffer solution,PBS)相比,视网膜的细胞排列有一定规则,组织水肿不明显;A组视网膜的VEGF水平明显低于B组,且都高于C组。结论血管抑素可明显降低糖尿病鼠视网膜组织的VEGF表达,能有效抑制糖尿病大鼠视网膜组织水肿及内部组织结构的改变。因此,血管抑素有潜在预防和治疗糖尿病性视网膜病变的价值。

Retinal Tissue Changes and the Expression of VEGF after Angiostatin Inhibition in Diabetic Rats

Abstract:Objective To investigate the inhibitory effect of Angiostatin on retinal neovascularization in vivo and the expression of vascular endothelial growth factor(VEGF).Methods One hundred and eight male adult Sprague-Dawley rats were induced for diabetes by an intraperitoneal injection of streptozotocin,then randomly divided into the model group(A)and normal group(B).And 36 SD rats as the normal control group(C).After intravitreal injection of vascular inhibition in rats,the retinal results were observed by HE staining and the expression of VEGF were evaluated by Western blotting.Results In the same time,the blood glucose of group A and group B were significantly higher than group C.Comparing the vascular inhibition injection in group A with PBS injectione in group B,there were regular arrangement and obscured edema of retinal cell tissue.VEGF level of retinal in group A was significantly lower than group B,both groups had higher level of VEGF than group C.Conclusions Angiostatin can obviously reduce VEGF expression,tissue edema and internal structural changes of retinal in diabetic rat.Consequently,angiostatin is potential to be used in prevention and treatment of diabetic retinopathy.
