海尔福口服液对自由基清除率影响的实验研究

摘    要:
目的研究海尔福口服液对羟自由基(.OH)和超氧阴离子自由基(O.-2)清除率的影响,以进一歩探讨其药理作用。方法根据Fenton反应原理,利用H2O2与2价铁离子Fe2+混合产生.OH,加入水杨酸产生有色化合物,在波长510 nm处有最大吸收峰,可比色测定。利用AP-TEMED系统产生O.-2,显色用分光光度法测定,加入抗氧化药物干扰后,系统产生自由基受抑制或清除,计算药物对.OH和O.-2清除率。结果海尔福口服液(浓度0.1 g/mL生药)、维生素C(浓度为1 g/L)和金银花对.OH清除率分别为(100.75±8.07)%,(99.42±0.14)%,(59.61±2.21)%。前两者比较,P>0.05,无显著性差异。前两者与金银花比较,P均<0.001,均有显著性差异。海尔福口服液对.OH和O.-2清除率分别是(100.75±8.07)%,(92.07±1.37)%,两者比较,P>0.05,无显著性差异。存放1个月、6 a、12 a的海尔福口服液样品对O.-2清除率分别为(91.72±1.38)%,(89.18±4.50)%,(70.37±1.24)%,存放12 a降低明显(P<0.01);维生素C作为标准对照,不同浓度对O.-2清除率r=0.996 7,有直线关系。不同浓度海尔福口服液对.OH清除率r=0.999 0,有直线关系。结论海尔福口服液对O.-和.OH有明显的清除率,是其药理作用之一。

Abstract:
Objective It is to study the influence of Haierfu oral liquid on the clearance of hydroxyl free radical(·OH) and superoxide anion free radical O·-2,and to make further investigation of the pharmacological effects.Methods According to Fenton reaction principle,H2O2 and ferrous ion mixed to produce ·OH,which added in salicylic acid to produce colored compound.The maximum absorption peak was at 510 nm wavelength and could measure with shade selection.O·-2 was generated according to AP-TEMED and the coloration was measured with spectrophotometry.After anti-oxidant drugs added to interfere with,the free radical produced by system was inhibited or cleared.The clearances of drugs on ·OH and O·-2 were computed.Results The clearance of Haierfu oral liquid(concentration 0.1 g/mL crude drug),vitamin C(concentration 1 g/L) and flos lonicerae japonicae on ·OH was(100.75±8.07)%,(99.42±0.14)% and(59.61±2.21)% respectively.The difference between the former two items was no significant(P>0.05),but the differences between the former two items and honeysuckle were both significant(both P<0.001).The clearance of Haierfu oral liquid on ·OH and O·-2 was(100.75±8.07)% and(92.07±1.37)% respectively and the difference between the two items was no significant(P>0.05).The clearance of the samples of Haierfu oral liquid stored for 1 month,6 a and 12 a on O·-2 was(91.72±1.38)%,(89.18±4.50)% and(70.37±1.24)% respectively and the one stored for 12 a lowered obviously of all(P<0.01).As standard control,the clearance of different concentration of vitamin C on O·-2 had straight line relationship(r=0.996 7).The clearance of different concentration of Haierfu oral liquid on O·-2 had straight line relationship(r=0.999 0).Conclusion Haierfu oral liquid has obvious clearance on ·OH and O·-2,which is one of its pharmacological effects.
