摘    要:目的 探讨多糖生物膜(PSBM)作为角膜内皮细胞培养及移植载体的可行性。方法 通过PSBM的不同制剂给予大白鼠口服,悬浮培养仓鼠肾(BHK21)细胞及兔角膜层间植入,观察其毒性、生物相容性及可吸收性。结果 食用含多糖微颗粒的饲料60d后,大白鼠无肝肾功能及组织学改变;PSBM微载体悬浮培养72h后,细胞密度可增值3倍以上;角膜层间植入PSBM后2~3个月时,植人物基本吸收。结论 PSBM属安全无毒物质,具有很好的细胞亲和性、生物相容性及可吸收性,是CE细胞培养及移植的良好载体。

Abstract:Objective To study the possibility of polysaccharide biomembrane ( PSBM ) as a carrier in cultured and transplanted corneal endothelial ( CE) cells. Methods ( 1 ) BHK21 cells were cultured suspendly on the polysaccharide microcarrier in RPMI1640 medium supplemented with 10% fetal bovine serum. (2) The PSBM was placed in rabbit' s corneal stroma. Results (1) The concentration of HBK21 cells increased three-folds at 74 h after culture. (2) PSBM in corneal stroma induced slight neovascularization along the wound and disappeared gradually at 2-3 postoperative months. Conclusion PSBM seem to have better biocompatibility and may be an excellent carrier for CE cells' culture and transplantation.
