巢蛋白在已分化的神经干细胞中表达时程的研究

摘    要:
目的 探讨神经干细胞分化过程中巢蛋白(nestin)表达的时程变化。方法 在细胞分化的不同时间进行免疫荧光染色。结果 在神经干细胞分化早期,巢蛋白表达仍呈阳性。随着分化的神经细胞、星形胶质细胞或少突胶质细胞逐渐发育成熟,巢蛋白表达渐渐减弱,到第7周时巢蛋白表达完全消失。结论 巢蛋白表达不仅是神经干细胞的重要标志,同时也出现在神经干细胞分化的早期,而且随着细胞逐渐分化成熟,其表达显著下降。体外培养情况下至分化的第7周即完全消失。

Abstract:
Objective To investigate the temporal expression of nestin in differentiated neural stem cells (NSCs).Methods Immunofluorescent staining was performed at different time points after differentiation.Results At early stage after differentiation,there was still nestin expression.As cells differentiated more and more mature,the expression of nestin was dropped gradually until completely disappeared at the end of the 7th week.Conclusion Nestin expression is an important marker of not only undifferentiated NSCs,but also NSCs in their early stage of differentiation.As the cells maturated,Nestin expression is decreased remarkably until completely disappear at the end of the 7th week.
