Caveolin-1蛋白在急性白血病中表达的研究

摘    要:目的探讨急性白血病(AL)初次诱导化疗缓解前后caveolin-1蛋白表达的变化及Caveolin-1蛋白的表达与AL分型的关系。方法采用流式细胞术检测65例AL患者及10例正常对照组Caveolin-1蛋白的表达。结果65例初治组AL患者表达Caveolin—1蛋白的阳性细胞率(2.31±3.62)%、平均荧光强度(69.88±21.17),分别较10例正常对照组的阳性细胞率(77.50±1.76)%、平均荧光强度(289.87±7.92)明显下降(P〈0.01);38例AL患者诱导化疗缓解后表达Caveolin-1蛋白的阳性细胞率(3.03±3.87)%与初治组比较无明显差异(P〉0.05),但平均荧光强度(152.74±25.81)较初治组明显升高(P〈0.01)。各种类型的急性白血病表达Caveolin-1蛋白的阳性细胞率、平均荧光强度无明显差异(P〉O.05)。结论急性白血病的Caveolin-1蛋白表达水平较正常人明显下降,且AL初次诱导化疗缓解后Caveolin-1蛋白的表达较初治时有所恢复.提示Caveolin-1蛋白的高表达可能与AL患者预后有关.

Abstract:Objective To investigate the variation of the expression of Caveolin-1 protein in leukemnia before and after the first chemotherapy,and the correlation between the expression of Caveolin-1 protein and the types of acute leukemia. Methods To detect leukemia ceils from bone marrow of 65 acute leukemia patients and hematopoietic cells from hone marrow of normal persons by the flow cytometry.Results The positive rate (2.31± 3.62)% and mean fluorescence intensity(69.88±21.17) of 65 acute leukemia cells of the untreated group(65 cases),which express Caveolin-1 proteins,were respectively lower than the positive rate(77.50±1.76)% and mean fluorescence intensity(289.87±7.92) of the normal control group(10 cases)(P〈0.01); The positive rate(3.03±3.87)% of acute leukemia cells of the remission group(38 cases) is not different from the untreated group's(P〉0.05),but the mean fluorescence intensity(152.74±25.81) of the former is higher than that of the latter(P〈0.01). The positive rate and mean fluorescence intensity of any type of acute leukemia cells were not respectively different from those of the other one(P〉0.05).Conclusion The expression of Caveolin-1 protein of acute leukemia cells is much lower than the hematopoietic cells',and its expression of the remission group after the first chemotherapy is higher than untreated group's to some extent,suggesting that high expression of Caveolin-1 protein is a good prognostic factor for acute leukemia.
