川芎嗪对烧伤大鼠心肌损伤保护作用的实验研究

摘    要:目的 探讨川芎嗪对烧伤大鼠心肌损伤的保护作用及可能机制。方法 采用30%体表面积Ⅲ度烫伤大鼠模型,64只Wistar大鼠随机分为烧伤组和川芎嗪组。另取8只大鼠作为伤前对照。于致伤前及伤后3、6、12和24h检测血清心肌钙蛋白T(cTnT)、心肌组织超氧化物歧化酶(SOD)含量变化;Westernblot检测心肌组织热休克蛋白70(HSP70)的表达。结果 大鼠严重烧伤后3h和12h心肌组织中HSP70蛋白的表达显著升高,血清cTnT含量显著升高,心肌组织中SOD活性显著降低(P〈0.01)。与烧伤组相比,川芎嗪组心肌组织中HSP70蛋白的表达明显升高,血清cTnT含量显著降低,而心肌组织中SOD活性显著升高(P〈0.05或P〈0.01)。结论川芎嗪对大鼠严重烧伤后早期心肌损伤具有一定的保护作用,其保护机制可能与HSP70诱导表达增强有关。

Abstract:Objective To investigate the protective effects and mechanism of tetramethylpyrazine(TMP) on myocardial injury following severe burn.Methods A total of 64 Wistar rats with 30% total body surface area(TBSA) fullthickness burn were randomly divided into two groups: burn group and tetramethylpyrazine treatment group.Another 8 Wistar rats were used as normal control.The levels of serum cardiac troponin T(cTnT) and the content of myocardial superoxide dismutase(SOD) were determined at 0,3,6,12 and 24 hours postburn.Samples of myocardium were taken for the determination of HSP70 expression by Western blot analysis.Results HSP70 expression in myocardium increased significantly at 3 and 12 hours after severe burns.Serum cTnT contents were all increased significantly,SOD in myocardial tissue decreased significantly(P<0.01)postburn.In the TMP treated group,HSP70 expression in myocardium and SOD concentration in myocardial tissue were increased significantly,serum cTnT level decreased significantly(P<0.05 or P<0.01)postburn compared to the Burn group.Conclusion TMP has protective effects on myocardial injury in burned rats and the mechanism might be closely related to enchancing expression of induced HSP70 in myocardium.
