摘    要:目的 检测食管鳞癌组织中F-box蛋白4(FBX4)和端粒重复序列结合因子1相互作用核蛋白2(TIN2)mRNA表达,并分析其与食管鳞癌相关临床指标的关系.方法 收集110例明确诊断的食管鳞癌中心组织及其30例癌旁组织,采用SBYR Green实时荧光定量PCR方法检测FBX4和TIN2基因在食管鳞癌中心组织和癌旁组织mRNA水平的表达,并分析FBX4和TIN2 mRNA表达临床相关指标的关系.结果 FBX4 mRNA在食管鳞癌组织中的相对表达量为0.24±0.43(n=110),而在肿瘤旁组织中为0.40±0.27(n=30),两者差异具有统计学意义(P=0.008);食管鳞癌组织中TIN2 mRNA的相对表达量为1.59±1.19(n=101);肿瘤旁为2.54±1.89(n=30),两者差异具有统计学意义(P=0.000).TIN2 mRNA表达在临床各参数不同分组内均未发现有统计学差异(P>0.05),而FBX4 mRNA表达虽在不同性别、年龄、钡餐分型、临床分期和肿瘤长度中均没有统计学差异(P>0.05),但在不同病理分型中存在统计学差异,即相对于高分化鳞癌,低分化和中分化的鳞癌组织中的FBX4 mRNA表达增加,差异具有统计学意义(P<0.05).FBX4和TIN2在肿瘤组织中的表达呈正相关(r=0.671,P=0.000).结论 FBX4和TIN2 mRNA在食管鳞癌组织中均表现表达增高,且存在直线相关关系,提示FBX4和TIN2可能在食管癌的发生发展中具有重要的临床意义;肿瘤分化程度与FBX4表达相关,而与TIN2不相关,表明肿瘤分化还与FBX4介导的泛素化相关.

Abstract:Objective To detect the expressions of TIN2 and FBX4 mRNA in esophageal squamous cell carcinoma, and analyze the relationship of esophageal squamous cell carcinoma associated with clinical indicators . Methods 110 cases of esophageal squamous cell carcinoma and 30 cases of its adjacent tissues were collected , using SBYR Green fluorescent quantitative PCR assay to detect the mRNA expression of FBX 4 and TIN2 genes in esophageal squamous cell cancer, and analysis the relationship between the mRNA expression of FBX 4 and TIN2 and clinically relevant indicators. Results The relative expression of FBX4 mRNA in esophageal squamous cell carcinoma was O. 24± 0. 43 (n = l IO), while in its adjacent tissues was 0. 40 ± O. 27 (n = 30), the difference had statistical significance(P =0. 008 );the relative expression of TIN2 mRNA in esophageal squamous cell carcinoma was 1.59 ± 1.19 ( n = 101 ) ; in its adjacent tissues was 2. 54 ± 1.89 ( n = 30 ), the difference had statistical significance ( P = 0. 000 ). TIN2 mRNA expression and various clinical parameters had not found significant difference (P 〉 0. 05 ), while FBX4 mRNA expression, although with gender, age, }~arium meal type, clinical stage and tumor length were not significantly different ( P 〉 0. 05 ). However, with the pathological type, there was a significant difference, which was compared with well-differentiated squamous cell carcinoma, FBX4 mRNA expression was significantly increased in poorly differentiated and middle differentiated squamous cell carcinoma tissues , with statistical siguificanee ( P 〈 0. 05 ). Conclusions FBX4 and TIN2 mI{NA in esophageal squamous cell carcinoma showed increased expression, and there is a linear correlation between them , suggesting FBXa and TIN2 in esophageal cancer development may have important clinical significance ; degree of tumor differentiation eonelated with FBX4 expression but rmt with TIN2 suggests tumor differentiation may be related with FBX 4 that mediated ubiquitination.
