鳖甲煎丸大黄虫丸联合华蟾素预防实验性肝癌的研究

目的观察鳖甲煎丸、大黄虫丸及华蟾素对肝癌形成的抑制作用。方法将大鼠随机分为正常组、病理组和治疗组,除正常组常规喂养外,余2组均以0.05%2-乙酰氨基芴饲料喂服诱发肝癌,同期治疗组予鳖甲煎丸和大黄虫丸口服及华蟾素腹腔注射,实验35 d后观察大鼠生存及肝功能、肝组织病理学和PCNA阳性表达等指标变化。结果病理组和治疗组ALT、AST、ALP水平均升高,A/G值降低,肝组织学检查可见癌结节形成,PCNA指数明显增高。治疗组肝功能损害程度和肝癌结节形成数目、PCNA指数、大鼠死亡数均明显低于病理组。结论鳖甲煎丸、大黄虫丸及华蟾素联合应用可抑制2-乙酰氨基芴诱发大鼠肝癌的发生,减轻肝功能的损害,其作用可能与抑制癌前变细胞DNA复制及细胞增殖有关。

Study of Biejiajian pill and Dahuang Zhechong pill combined with Huachansu in preventing experimental liver cancer

Objective It is to observe the inhibition function of Biejiajian pill and Dahuang Zhechong pill combined with Huachansu on the formulation of liver cancer.Methods The rats were randomly divided into normal group,pathological group and treatment group.All the groups were fed with 0.05% 2-AAF to induce liver cancer except normal group.The treatment group was given Biejiajian pill and Dahuang Zhechong pill taken orally and Huachansu with intraperitoneal injection.The indexes of survival,liver function,liver tissue pathology and PCNA positive expression of the rats were observed after experiment for 35 d.Results In pathological group and treatment group,the levels of ALT,AST and ALP all rose up, the A/G value lowered down,the formulation of cancer tubercle could be found with examination of liver histology and the index number of PCNA was obviously increased.But the damage degree of liver function,the numbers of liver cancer tubercle,index number of PCNA and the dead rats in treatment group were obviously lower than those in pathological group.Conclusion Biejiajian pill and Dahuang Zhechong pill combined with Huachansu can inhibit the formulation of liver cancer induced by 2-AAF in rats and can relieve the damage of liver function.The mechanism may be related with DNA replication and proliferation of precancerous lesion cells inhibited.
