摘    要:目的对深圳市首例人禽流感病例进行实验室确诊,并研究其携带病毒关键基因的遗传特性。方法采用实时荧光RT-PCR方法检测高致病性禽流感病毒(H5N1)核酸,采用血凝抑制方法检测患者血清抗高致病性禽流感病毒的抗体滴度变化,利用MEGA3.1软件分析患者携带病毒HA、NA基因的进化关系。结果成功确认了深圳市首例人禽流感病例,患者携带病毒为H5N1高致病性禽流感病毒,HA、NA基因进化关系比较特殊。结论深圳地区禽鸟中可能携带高致病性禽流感病毒,具有潜在感染人类的可能。

Laboratory test of first patient infected With avian influenza virus in Shenzhen City

Abstract:Objective To confirm the first patient infected by avian influenza virus in Shenzhen and observe the genetic characterization of the highly pathogenic virus isolated from the patient. Methods Real time RT - PCR method was used to detect the virus( H5N1 ) nucleic acids and the serum titer between recovery phase and acute phase was determine by HI assay to confirm whether the patient was infected by avian influenza viurs( H5N1 ). Phylogenic tree of HA and NA gene was respec- tively constructed by using MEGA 3.1 software to analyze the evolutionary relationship between Shenzhen avian virus and the others. Results The case was confirmed to be the first patient infected by high pathogenic avian virus( H5N1 ) in Shenz-hen. Conclusion The (poultries) fowls and birds in Shenzhen may, to a certain extent, carry highly pathogenic avian influenza virus, which may patentially transfect from fowl to human.
