不同抗凝剂对乙肝表面抗体检测结果的影响

目的验证不同抗凝剂对试验结果的影响。方法采用确定HBsAb阳性(其中包括临界值阳性)和阴性两组志愿者,阳性对照组50例,阴性对照组10例,再对每一位志愿者同时采集血清和不同抗凝剂处理的血浆样本,用安图化学发光表面抗体定定量检测试剂测定不同抗凝剂处理的样本的表面抗体含量,比较差异。结果用肝素作为抗凝剂的血浆样本与血清样本检测结果的偏差阳性值不超过±10%,相关性r=0.9954。用枸橼酸钠和EDTA作为抗凝剂结果均超出了以上范围。结论本试剂盒的样本类型可以是血清或血浆,抗凝剂建议使用肝素,用枸橼酸钠和EDTA作为抗凝剂的血浆不能作为本试剂盒的检测。

DIFFERENT ANTICOAGULANTS FOR QUANTITATIVE DETERMINATION OF ANTI-HBS CONCENTRATION IN HUMAN SERUM

Objective To analysis the effects of different anticoagulants on clinical test results for quantitative determination of anti - HBs in human serum. Methods Take the samples which are determined as the anti - HBs positive including the critical pos- itive value and the anti -HBs negative from two groups of volunteers. Negative samples group has 10 cases, while the positive samples group 50 cases. Then for each volunteer, collecte serum and plasma samples treated by different anticoagulants. Then determined the anti - HBs concentration with Autobio CLIA Test Kits. Results In the positive group the deviation of anti - HBs concentration between the plasma sample treated by Heparin sodium and serum is less than 10%, correlation is 0. 995 4,in the negative group less than ± 10% ,while the deviation between the plasma samples treated by EDTA - Na/K or sodium citrate is more than 20% or ± 20%. Conclusion The samples of Autobio CLIA Test Kits can be serum or plasma, and the suggested anticoagulant is Heparin sodium, samples treated by EDTA- 2Na/K（ 1.5 mg/ml）or sodium citrate should not be used.
