抗CEA单克隆抗体在卵巢癌病人体内的血药动力学研究

摘    要:
研究了~(131)I-CEA C50在22例卵巢癌病人放射免疫显像中的血药动力学特性。~(130)I-C50的血半清除时间平均为T1/2α=24.8h,T1/2β=40.5h(n=12)。在静脉给药后28h内约有给予放射性总剂量的50%由尿排出。循环血液中标记抗体主要在血浆中。卵巢癌病人在接受静注标记抗体后2~3周即可产生人抗鼠抗体,在体内维持至少9个月以上。二次显像应用时,抗鼠抗体的产生影响了二次显像效果,限制了鼠源性单抗的反复应用。

Abstract:
The biokinetics of ~(131)i-labelled monoclonal antibodies againstCEA have been examined in 22 ovarian cancer patients who were receivinglabelled antibody in diagnostic immunoscintigraphy trials.A hi-phasic declineof radiolabelled preparations was measured with initial half-lives 24.8 hoursand then with 40.5 hours in second half-lives.Blood-borne radioactivity wasassociated predominantly with plasma rather than cellular elements.Urinaryexcretion of radioactivity was about 50% of injected dose in the first 28 hours.However,anti-mouse-IgG antibody(HAMA)detected 14 days after admini-stration of radiolabelled antibody and was present for at least 9 months,and has implications for the efficiency of repeated image studies.
