肉苁蓉总苷对D-半乳糖脑老化模型小鼠海马超微结构的影响

摘    要:目的观察肉苁蓉总苷(GCs)对D-半乳糖(D-gal)脑化模型小鼠海马超微结构的影响。方法NIH小鼠120只,随机分为6组:正常对照组(NC)、模型组(D-gal)、VitaminE组、GCs组分小、中、大剂量组(mg.kg-1.d-1)31.25,62.5,125],每组20只。除NC组外,在各组小鼠皮下注射D-gal(50mg.kg-1.d-1);NC组皮下注射NS,D-gal组和NC组给予等容积饮水灌胃,VitaminE组和GCs组灌胃给药,每日1次,均连续皮下注射和灌胃50d,取小鼠海马CA1区进行电镜观察。结果GCs小、中剂量组及VitaminE组的海马CA1区神经细胞核形不规则,细胞器明显减少,染色质有不同程度的凝集;GCs大剂量组海马CA1区神经细胞核圆,细胞器增多,染色质分布均匀,与NC组的形态结构较接近。结论GCs对D-gal脑老化模型小鼠海马超微结构的保护作用呈剂量效应关系。推测GCs可能是通过抗氧化机制起到延缓衰老和防治老年性痴呆的作用。

Abstract:Objective To observe the influence of glycosides of cistanche (GCs) on the ultrastructure of hippocampus of the D-galactose induced brain aging model mice.Methods 120 NIH mice of every stage were randomly divided into six groups: normal control group (NC), model group (D-gal), VitaminE group , GCs groups which was divided into lower dose, middle dose and large dose(mg.kg-1d ~1) 31.25, 62.5, 125]. Every group had 20 mice. Except for NC group, other groups were given hypodermic injection of D-gal (50mg.kg~-1.d~-1). NC group was given hypodermic injection of NS. D-gal group and NC group were given oral administration of water.Vitamin E group and GCs groups were given oral administration. Mice of various groups of every stage were repeatedly given hypodermic injection and oral administration one time every day , and the tissue of hippocampus CA1 region of those mice were examined with electron microscope after 50 days. Results The form of cell nucleus was changed , cell organelles reduced obviously and chromatin of cell nucleus appeared various degree of agglutination on hippocampus CA1 region in lower dose , middle dose of GCs groups and Vitamin E group. The cell nucleus was round , cell organelles was increased and chromatin of cell nucleus was well distributed on hippocampus CA1 region in large dose of GCs,which was similar to NC group. Conclusions With increase of dose , protective effect of GCs was increased for ultrastructure of hippocampus of the D-galactose induced brain aging model mice. It indicated that GCs could slow decrepitude and has effect of prevention and treatment of aged dementia , the mechanism of which may related with antioxidation .
