兔脊髓缺血再灌注脂质过氧化和细胞凋亡变化及川芎嗪对其的影响

摘    要:
目的:探讨川芎嗪(TMP)对兔脊髓血再灌注(IR)脂质过氧化及细胞凋亡的影响。方法:参照Zivin法建立脊髓IR模型,将兔随机分为假手术组、模型组和TMP处理组。检测脊髓组织超氧化物歧化酶(SOD)活性丙二醛(MDA)水平,分析脊髓神经细胞凋亡指数和凋亡相关蛋白Bcl-2,Bax表达水平,评价再灌注后24h,48h后肢运动神经功能。结果:TMP可明显提高脊髓SOD活性,降低MDA水平,可通过上调Bcl-2,下调Bax的表达,抑制缺血再灌注所致的脊髓神经细胞凋亡;改善缺血再灌注动物的后肢运动神经功能。结论:TMP对脊髓IR有保护作用,其作用与减少神经细胞脂质过氧化损伤和抑制神经细胞凋亡有关。

Abstract:
Objective:To study the effects of Tetramethylpyrazine(TMP)on lipid peroxidation and apoptosis after spinal cord ischemia reperfusion in rabbits.Methods:the spinal cord ischemia in rabbit was established by method of zivin,New Zealand white rabbits(n=27)were randomly divided into three groups:sham group,model group and TMP group.At the 24h,48th hour after reperfusion,the locomotive function of posterior lamb was assessed.We examined the expression of Bcl-2,and Bax by immunohistochemical staining,the apoptosis of nerve cell by TUNEL staining and measured the spinal SOD and MDA.Results:I/R prlduced a significant decrease in neurological scoring and TMP halved the decrease,as well as raises the activity of SOD and decrease the level of MDA.And it depresses spinal cord apoptosis caused by ischemia reperfusion through up-regulating Bcl-2 expression and down-regulating Bax expression.Conclusion:TMP has protective effects against spinal cord ischemia reperfusion injury,and its mechanism may be related to scavenging of oxygen radicals and inhibiting apoptosis.
